First time agar user here. Just a few procedural questions before I get started...

Okay, the 20 agar dishes I ordered come in a pre-sterilized sleeve. I'm afraid that by taking them out of the sleeve, I will compromise their sterility. Because of that, I feel like it would be best to pour my hot and freshly sterilized agar mix into all 20 dishes at the same time in my glovebox, then wrap in parafilm, then place back in the plastic sleeve they came in until they cool and solidify. I don't plan on inoculating all 20 at the same time with LC or a spore syringe, so I plan on just using 2 or 3 right away, and just keeping the other 17 or so in a plasitic bag in the fridge for sterility's sake. Then, whenever I need to do an agar transfer, I just take them out of the plastic bag and use each dish as necessary.
Is this the right way to go about handling my agar dishes?

Well for storage of blank poured plates, you really should store them in the fridge. Other than that, yes you should be good to do that. The ones in the fridge should get parafilmed, double bagged and left in a thermal lunch box to reduce condensation caused from temperature fluctuations.

You been around 5 years and you're just starting agar? Sheesh. You're really gonna be kicking yourself in the ass once you get started because you didn't start it sooner.

I know. I pretty much exclusively used liquid culture started from multispore. I had a very high contam rate, so I decided to give agar a shot since I've been told that it really is the way to go.

Spores to lc is probably the worst way to grow.
You won't be disappointed in your transition to agar.

I love agar. I also like to use clean agar wedges to inoculate my LC. Killer

One more question...Should I seal the petri's with parafilm once I finish pouring them, or should I pour them, place them back in the plastic sleeve and only seal with parafilm once they have finished solidifying? It would seem to me that it would be best to wait until they solidify, as I could accidentally spill some of the agar while sealing them.
