Inflating spawn bags

Hey all

Do you folks do this? I usually inflate my bags in the flow from the hood before sealing them up. Its how I learned.
But Im working on my technique trying to conquer mold problems and I feel like if I omit this its just less time for contams to slip in if Im clumbsy or something.
I mean I dont think you can do this is you work in a SAB anyway right?

Omitting the inflation step would encourage anaerobic environments, and therefore would be counter-productive.  Your sterile technique, dirty cultures, spawning in a dirty lab, or improper sterilization of substrate are your issue(s) if you are experiencing mold during colonization of grains in bags.

Try upping your sterilization duration.  Run your hood for longer before entering the lab.  Intensely clean your lab.  Hang plastic sheeting on all surfaces of your lab.  Start with new cultures.  Wipe all tools, and media thoroughly with an alcohol soaked paper towel immediately before placing them in the direct downstream of the flowhood.

Yea Loaf Ive been working on all of the above really, been really determined to lose 0 tubs

How do people manage to inflate in still air box?

No one really does bags too much in a sab.

The bags will semi self inflate if you hang them or otherwise cause an imbalance of pressure in them.

I've flipped mine upside down the day I make them. It gently and slowly forced the bag apart making it pull air through the filter. I wouldn't recommend this for anything larger than the .02um filters though.

Inflating bags inside of a still air box is like asking for the dehydrated water to be fetched.  :lol:

Grain transferring jars to bags inside of a still air box is an uncomfortable, time consuming, back breaking, studious task.

Inoculating bags with grains is EASIEST when done in front of the flowhood.  If you want to inoculate bags without a hood, the easiest way to do that is to pre-seal bags before sterilization and inoculate them with a liquid inoculant of sorts.

Ahh yea thats what I was confused about I was thinking "Dont people use spawn bags in SAB?"
Was wondering how that works if you cant fill them back up with air
