cloning in grain jar, can i shake? and other questions?

So i took a tissue from a big fast growing fruit and drop it in a jar smaller than a half pint...it took a few days to start colonizing and now it reached 35%.

1-can i shake it?
I want it to colonize fast to avoid contamination!

2-i would like to use the same strain in the future, how can i achieve this, should i g2g to 1 or 2 quart jars and use em as a master jars g2g to others when i want and more g2gs and store the original 2 in the fridge ?
like this:
tissue in the half pint--->g2g to 2 quarts--->g2g to a jar when needed----use this jar as a master g2g to other jars.

1- you can shake.

2- If you want to preserve a good performing culture, look into making master slants.

You shouldn't have dropped cloned tissue directly onto sterilized grain. You should have taken the clone to agar, cleaned it up, then dropped healthy mycelium onto sterilized grain.

I would worry about bacteria problems further down the road once you start expanding.  That tissue sample probably isn't 100% clean, so it might be doing well now but over time/transfers the bacteria has time to 'establish' itself.

That sounds good though, put that half pint to 2 quart jars, save one, expand the others, and see what happens.  Probably be fine for one or two transfers.

well if it didnt contaminate then why not just use grain!
millet spawned to CVG monotub

if you grew the mushroom in open air I wouldnt put it right into grains.
And I would especially not save those grain jars for use as masters later.

Fruiting is not a sterile process. Just because you don't see bacteria, spores, etc on your fruits doesn't mean they aren't there. You've effectively transferred dirty tissue to clean, sterilized grain. If it doesn't contaminate I would be really surprised.

But PF cloned using grains before! Yes it took him alot of jars but it worked and with other people too...and yea of course agar is way better but i cant work with it for now.
i transferred the tissue in a sterile condition but yea they could still be contam!
what if the tissue 100% colonized the jar without any noticeable contam?

Thats why i want to shake it so i can make things go a little bit faster but i dont know if you can shake a jar with a tissue inside it!

I would just go for it if they look pretty healthy after shaking that half pint, worst thing that happens is you lose some time and grains.  I wouldn't do it if you plan on doing more than a tub or two with it though. 

After the half-pint fully colonizes, shake it up again and see how fast it recovers, should recover quickly if its relatively bacteria free.

When you go for spawning to bulk for the tub, just use a high ratio so it colonizes quickly incase you have some bacteria buddies riding along.  (which you probaly do)

Well i just thought that the tissue is more prone to contam if its not surrounded by myc...well iam just learning! Lol

I go from clone to grain all the time. Clones / pins taken straight to agar and then healthy ones dropped directly to grain. Or clone tissue / pins dropped directly onto grains. Plenty of people have high success taking clone tissue directly to grains.

Of course it's more risky.  But for a home grower it's not an invalid way to get clones growing.

Try looking into liquid cultures, but honestly, agar is the way to go. Even if your jar is contamed, you can still save that clone using agar!

cron taught me to simply stab the pins on agar plates with a flamed syringe needle, rip him up and drop wherever you want. real easy!
