question about incubation / phase after inoculation
    #21925340 - 07/10/15 04:44 PM (8 years, 6 months ago)

Hey guys, after I inoculated the jars, do I need to leave them sealed, covered with aluminum foil or with the lid exposed with a hole and the filter? .

I think my biggest question is : does the mycelium need gas exchange (c02/02) after they are put in a jar or they need to be isolated to they don't get contamined?

Thanks,
Daniel

The foil is there so water doesn't drip into your jar during pressure cooking. If you are using a synthetic filter disk, are using poly fill or if you have self healing injection ports, remove the foil.

Are you doing PF? If so you can leave it or remove it. If you leave it it might be more likely to stall. If you take it off there's a higher chance of contams although it should not be by much if you did your dry verm layer correctly.

No matter which method you are using, take the foil off. It needs to breath, plus I would speculate that the foil could trap moisture and create a bacteria problem.

I'm still new, so take it for what it's worth... It's my understanding that in PF tek... You should take off the foil, If you are using Verm or a filter, you should be fine. They need to breathe.:crazy2:

Leave the inoc ports uncovered for gas exchange. They need gas exchange or else it will slow the growth by a lot. Also you don't need to incubate the jars, just put them on a shelf and don't fuck with them too much.

I had this same question about whether or not you need to have the ports covered. My response is the same response many trusted cultivators gave me. So use the advice.

It's being stored/ incubated in the SAB box which I carefully hygenized with alchohol and lysol using gloves, the bottles were also hygenized from outside and the box is closed

Totally unnecessary. I keep mine in a tote that has the lid off just so I can move them all at once when I need too. The dry vermiculite layer you put on top serves as a contamination filter. So in other words as long as your not fucking with the jars making that layer move around you have nothing to worry about. The thing that gave me contam problems in my jars was I left the tape on for the first 5 or 6 days and it kept in too much moisture which caused mold/bacteria growth issues.

I just checked the jars and they are mostly half colonized.
Do I shake them?

some pics:

Another question.

I have coco coir and frozen cooked grains and I have plans to do a bulk method. Do I still have a big contamination risk after these jars are colonized and placed in the fruiting chamber? any suggestions on the method to use?

Thanks,
Daniel

Oh you're using grains. Well once the jar is fully colonized the odds of contamination are greatly decreased.

even if you add more substrate when moving to the big container? or I'm not supposed to do that and just add coco coir?

I think I might be confused about this.
I'm not supposed to add extra substrate (fungus food) after the jar process, but just casing (coco coir, vermiculate) correct?

If you add more substrate then you must wait for it to colonize the new substrate.

I don't see vermiculite in those jars, it looks like some sort of grains or something to me  if it is verm then he got some real chunky shit lol

I've never heard of mixing grains with verm. Not to say that it's a bad idea to do so, but I've only ever seen people inoc jars that were only grains or only wbs as far as I know they didn't mix anything with them. Hopefully a grower more experienced with grains can weigh in on this. And if you got the mixing info from a tek on here then it's probably correct.

I think verm could be beneficial actually, but it may make the grains difficult to get out of the jar. I've never seen any trusted tek that talks about mixing grains with verm. You might be ok. If you haven't already, shake the jars and then maybe do it one more time before 100%. If the grains and verm form a solid mass then you may have to break the jar to get it out? Looks healthy otherwise.
