Wish me luck! I followed the tek's pouring directions keeping everything but the neck of the bottle from hovering over the top of the open dish and setting the stack promptly on top of it. I guess the next step is to re-bag em and give them a couple of days to see if they grow anything in them before testing the syringe on several?

I'm basically cross posting from the recipe thread:
Sorry I cannot figure out how to make a new post but only reply to a previous post.
I'm trying to troubleshoot why I'm having difficulty growing mycelium on agar (cubensis). I thought it was my spores so I bought new spores. I'm still having zero growth (no bacteria either for what it's worth lol).
I followed the 10g ELME/10g agar/1g yeast recipe. I used nutritional yeast. The ELME I used was this

Did I screw up? Is there something else I should have used? It's been 7 days with new spores and still no growth. Thank you

Spores can take a while, i usually wait 21 days before swabbing new plates
Lower agar% makes softer plates that germ better. Skip the nutritional yeast, otherwise your recipe sounds good for 500ml. Drop to 8g agar agar if you dont see anything in a couple more weeks.
A couple drops of sterilized water on the other plates might help too, dryer old spores take the longest. iβve seen germ after a month..

gives me hope for my starry nights^^ there like in week 2 and don't see anything considerable to growth. But as usual, I am optimistic until proofed otherwise :3

Second time doing this tek, the first time was a smashing success.
This time I did things slightly differently as an experiment. After PC'ing my agar, I took it directly out of the PC and submerged it in a bucket with an immersion heater set at 47c and let it "cook"/cool down for about an hour before pouring so it'd be just the right temp. The media bottle was fully submerged.
I just finished pouring my plates, and it went fine for the most part, except a few plates ended up with small chunks of agar that seemed to have solidified. Not enough to cause any issues I'm guessing, but still unexpected.
Anyone have any idea what might have happened? I would have thought that an immersion cooker would be the perfect way to keep agar at temp, similar to what actual labs presumably do, but it didn't work out so great for me. Did I maybe let it sit at 47c for too long?

I use a thermostatic water bath, the same as used in labs, but set it to 60Β°C. I would have the same problem starting at 47Β°C.

I plated penis envy January 28. It's almost March 28 and I don't have pins or knots or anything. Should I be worried or can I use this to transfer to grain? It doesn't appear to be contaminated

I think you are slightly misunderstanding the point of agar. You only want it to pin or knot if you are trying to clone it. If you are going to do agar > grain, you make a few transfers until you are sure it is clean growth, then let the colony grow out to about 2/3rds of the plate diameter and transfer to grain then. This process takes a few weeks.
You usually don't want to use a fully colonized plate to transfer to grain because the mycelium gets the signal to stop expanding because it has reached the edge.

Hi nice write up much appreciated!
I'm trying to get to the root of my agar issues and was hoping you could help?
I've been having problems with aerial mycelium and mycelium growing through the agar, not on the surface.
I've been using the MYA recipe:
10g agar 10g light malt extract 0.25g nutritional yeast. 500ml water
I've tried tweaking this recipe a few times on the advice of other forum members, but I'm still getting the same issues.
I didn't do the step of boiling my mixture before putting it in the pressure cooker and suspect this might be causing the issues. What do you think?
I've ordered powdered nutritional yeast as opposed to the flakes that I had been using, and a different brand of agar agar just in case. I'm going to try boiling my solution as I've said and try upping the nutritional yeast powder to 1g.
Any thoughts or insight would be greatly appreciated, and hopefully we can sort this on-going issue that I'm having.

I got into Agar recently and there is no reason Iβd put spore to grain ever again (unless I feel like playing mycelium roulette).
