Hello all, thanks for all the info I wanted to ask since my Agar seems to become quite solid, could this be on due to to long time in the pc? Bod writes “agar is sterilized in 20m at 15psi. I always usually go to 20m I would not go longer than 30-45m you risk caremelization of sugars in your media.” So I did about 25 min plus the time it took to vent properly (10-15 min). I used 8g Agar agar, 6g MAE and 0,4 liters of water.
The mycelium seems to have difficulties growing on the hard agar I produced, does anyone have any suggestions? Cheers

try using less agar-agar powder. Drop it down to around 7.5g instead of 8g next time and adjust it from there. But, 8g in 400mL of water is what's considered "standard", so you may want to rule out all other possibilities for why it's having trouble growing before pointing the finger at your agars consistency.
Are you just now germinating from spore print/syringe? or are you past that and making transfers? First signs of germination took 5 days on both cubes and pan cyans, so if that's the stage you're at you may need to just be more patient

Found this to be very helpful. My second attempt at growing is with some LC on 3 agar dishes. Makes sense to prep lots of dishes, and store the ones you don't use straightaway for later.
Will transfer the best looking growth to new dishes a few times to try and get as clean as possible growth, before i go to grain.

That’s what I had been thinking when I started agar. I would get a clean plate and then transfer to like five more to grow out for spawn jars. Then I tried g2g. So much better. Now I just keep a plate or two around, drop a wedge to like a quarter quart of seed then g2g that once it’s colonized.

Probably a dumb question, but what does 20g/L means?

I don't get it, so what is the formulas for these?
Grain Soak Water agar(save water from hydration of grain spawn add 20g/L Agar-Agar powder) BRF agar (20g/L BRF and Agar-Agar) MEA, malt extract agar. Use light malt extract for homebrewing at 20g/L YPD, yeast peptone dextrose agar, I buy this premade and use as is PDA, I just kind of wing this one every time I use it

For every liter of grain soak water add 20g of agar-agar
For every liter of water add 20g brf and 20g agar-agar
For every liter of water add 20g malt and 20g agar-agar
the formula is what you copied and pasted

I recently purchased a 7 lb bag of malt extract (it was cheaper per pound than the one pound bags). Unfortunately, when it arrived I realized that it's syrupy liquid...can it still be used effectively for agar? Should I just return it? If it can be used, any idea how much would be equivalent to dry malt extract? Ratios of malt extract liquid to agar, water, etc. Thank you in advance for any assistance.

Yes, also brown rice syrup works nice. That's sort of a pricey thing but actually good to have around for sweetening things.

Is it a solution if I make a biopsy on a beautiful and potent mushroom, clone it, and keep it as the master?
If it is, does it also mean that in theory I never have to make spore prints of the same strain again?
Clone it. It isn't a forever thing, but practically you are set.

Hey everyone!
First time posting and growing, not sure if it's the right place to ask, but I think my issue is related to my agar--
I followed bod's grain soak agar recipe to produce my agar plates-- 10g agar per 500mL of race horse oats water left over from boiling oats according to bod's oat prep guide-- 12 cups horse oats, 10L water, 30m to get oats to boil, 40 minutes at rolling boil, oats started splitting, etc. (I did not do a soak beforehand) I poured around 120-125 degrees, inoculated ultimately twenty plates with a shaken spore syringe and used a loop to zigzag across each once the plates solidified.
The issue is it's been about three weeks (two weeks for the first ten plates, a week for the second ten plates) and I've seen no signs of growth on any plates-- not even contaminants. Temperature is consistently around 65-68 degrees in my house and SAB.
Any ideas what could be wrong? Excessive condensation on the ceilings of the plates? Too nutrient-rich? The use of non-vented plates?
Any help would be appreciated

What's up BOD,
I'm a noob on my 3rd sleeve of petri dishes experimenting with agar. Definitely appreciate all your teks. But I'm sitting around waiting for my agar to cool again, so I figured I'd share my mistake so my fellow noobs might avoid it.
When tightening the lid on 500ml media jars after PC equalization...DONT tighten the lid like an animal. The lid contracts as it cools and gets a vice grip on the jar. I've got a monster blister as a memento. Reheating in boiling water is the answer, but that was lame.
Cheers!!

You adjust time accordingly. I'm sure 30 mins is plenty at 12psi for agar. Some folks I think run way overkill on liquids.
In fact 30 may be Overkill.
