How long sitting in the fridge before it's best to just make up another batch of agar? I made up 400 mL of LME agar and forgot about it. It's been sitting in the fridge for around 4-5 days.

Sterilize at 15PSI for 20m
Does this include the 10mins of venting with the lid on? Or is it 10 mins venting, then 20 mins sterilizing? Also do you start the sterilization timer when the weight rattles or when it’s first put on after venting? (Mine takes about 5 minutes to start rattling on high heat, after venting)

No that doesn't include vent time. So let it vent for 10-15 mins, then sterilize for 20 minutes at 15 psi. You don't start your 20 minute timer until it hits 15 psi

read the manual with your pressure cooker and look at the basics of pressure cooker operation and my pressure cooker tek. sterilization doesn't include the vent time. it doesn't even start until pressure is reached so not only after venting but after it reaches pressure as well. that's when the time starts.

Hi all!
I’m really new to this so please forgive any misunderstandings I’m just a bit confused, I tried out the grain soak water Agar recipe. First I thought this was the water from I got over from boiling the oats, but looking at my agar development (or lack mycelium growth would be a better description) 3 weeks later figured I did something wrong since there is no progress at all, no mycelium, no mold, nothing..did I use the wrong water for the recipe? since there is no step for soaking oats 12-24h before boiling them on Bods oat prep I just got confused and used the water I got over from boiling the oats. Re-do, do right?
The agar I got was also really hard, could this also affect the growth?
Again, I’m really new to this, trying to find my path through the jungle of TEKs )
Any help is greatly appreciated!

When I microwave my agar, I'll twist it just enough for it to loosen just a tad. Normally this let's out a brief, weak hiss. Probably a stupid question, but that's not opening it too much, is it?

Is there a good way to store LME? I have a 5 pound block of it that's as hard as a rock. It's so hard I can't even scrape off 8 grams.
When I buy some more I think I will put it in 8 gram packets in foil and keep them in mason jars. Should I freeze them?
One other question. Due to the problem above I PC'd 400 ml of agar mixture with 8 grams each of agar and BRF. There's quite a bit of sediment. I wanted to filter it before cooking but I couldn't find my house mates coffee filters. Should I pour this stuff or wait to get LME and make clear agar?

Air tight. Your agar is fine.

I was thinking doing a bunch of agar in media bottles and sterilize them all at once. Re-melting in the microwave as needed. Does anyone here do it as well?

I noticed this tek says
Quote:
then place into your pressure cooker, I like to make the water in the PC come up to the level of the agar in the bottle. this will make it take longer to hit pressure and cool down but will cut down on boil over.
And PC one says
Quote:
As long as half of your jar is above water there's sufficient contact surface for STEAM. STEAM is what does the sterilization you want your jars to have at least 50% of their surface above the water so that steam can transfer energy to the jars.
I'm guessing it doesn't make a huge difference correct?
I just threw some agar in with the water 1/2 way up the agar, let's see if I get boil over.
Update: Def boiled over, don't do this

Is there a rough formula by chance regarding the agar water bath? I know too much water will obviously pull too much heat out too fast and not enough will leave you waiting around. I like to work with like 2000ml of pouring at a time so these details get important.

No formula. Real labs have a heated water bath which will get the agar to perfect temp and keep it there. I start with hot water for the bath and when the temp hits pouring temp i assume the entire thing is homogeneous temperature and its worked for me that way.

It's not necessary, obviously, but if you regularly pour such amounts, I can recommend a home sous vide cooker, a.k.a. immersion circulator. I got mine years ago for cooking and it works wonderfully for keeping my agar bottles at just the right temp (4x500ml bottles in a large pot). Might be worth it, even if you don't cook (though you should try it for steaks!).

Noted. Not sure why I didn't think of that. Also a justified purchase to the woman.

Forgive me if I missed this, but for the 400ml is it one 25g packet of agar-agar and 8oz of light malt extract?
