Thank you, awesome TEK BOD! Is there a time limit on the grain soak water? I made way too much and have it stored in the fridge until I have a chance to make more agar.

Glass ones are a pain to work with and usually a few bucks a piece is a good deal.

I’ve had great success with your tek, thanks! Also recommended to a friend who failed with every previous recipe he tried, and he’s had great results with yours!

http://bioland-sci.com/index.php?main_page=product_info&products_id=1279 $0.13 is cheap enough to not mess with them, here’s the link for $65 case of 500 I found. Sorry for the annoyance

Yes, kind of the only way to store them unless you have a lot of tables

then place into your pressure cooker, I like to make the water in the PC come up to the level of the agar in the bottle. this will make it take longer to hit pressure and cool down but will cut down on boil over.
I understand this as filling the PC with water up to level C. Is that correct? Or rather B? Intuitively I would have filled up to A or even a bit lower.

Posted in another thread, but I've been doing heaps of bulk grows with unmod tubs and spore syringe to jars, all with great success and no contams beyond a few trich situations when the heat got to much in the room after a bunch of flushes. So I decided to try out some Pan Cy's and thought, hey let's try out some agar plates this time.
Used needle to agar plate via these methods and all my plates went mold on me within 4 days. I'm heaps particular on sterile environments in my SAB, yet still got contam. Day one the dishes had a bunch of white circles starting and then the next morning were all grey spots that only got darker. I just tried some new dishes to see if maybe it was just a fluke. Any thoughts? Followed all the instructions well, didn't know if maybe that strain is just hard to start in agar possibly.

Thanks for all the great work, Boddhisatta, I learned a lot from your threads. I have 20 petri dishes and I'm wondering how to best use them. How many should I inoculate right away, and how many should I save for transfers? Also, should I fill them all with agar right away, and store the rest wrapped in saran wrap? Basically, fill all with agar, inocululate about five, and save the rest?

Do I need to use glass petri dishes? Or is it ok to use plastic petri dishes? Could you provide links to amazon to proper dishes please?

Some use glass but buying petri dishes in bulk and either premixed MEA or PDA (dehydrated) by the 100g brings the cost of agar work down to the disposable level. 100 bucks for a year+ supply of agar plates makes it alot easier to just saran wrap them and throw them away when the contam or break or whatever. https://www.amazon.com/Celltreat-229694-Petri-Slideable-Sterile/dp/B014AFZ820/ref=sr_1_2?keywords=500+celltreat+petri+dishes&qid=1566807738&s=gateway&sr=8-2
