Bod, I'm using the same 23 qt Presto pressure cooker/canner and I see that you suggest filling the water up to where the agar is. Is that safe to do in this pressure canner? I just want to be sure.

I removed ratings even though I only had exactly one and it was +5. Realizing this system was in place gave me anxiety. Way too much pressure to be liked by everyone.

Oh and do you vent for the same amount of time?

Filling up to the agar level helps prevent agar boilover. It's just fine to do that in a canner.

Check your manual. It may suggest venting for longer.

Since the idea is to push the air out of the canner I would think the amount of time of venting needed would be lower since water takes up more volume and there is less air, BUT I totally went longer at 15 minutes or so. If you guys have some science to drop on me to refute that reasoning please lemme know.
Totally went ahead and did it with the Presto 23 qt just now so I can confirm a person won't die by adding more water. It actually reached pressure wayyy quicker than normal but then took way longer to depressurize at the end.

My directions are not unsafe. And i use the 23q presto with my directions just fine. Since you have no good way of telling if you vented enough i just opt to do overkill venting

I thought it was possible you were using a different pressure cooker for this, and even if you were I'm not sure I had anything to worry about. Pressure cookers/canners just scare the shit out of me. Regardless, appreciate the reply. Wouldn't be here if it weren't for your teks.

How long and at what temp do you find it takes to properly oven dry your foil wrapped pyrex petri dishes? TIA, and thank you for this well written tek.

Don't put them in the oven wet ever. They need to be washed and cleaned really well then dried before oven sterilization. The time it takes is in the directions for dry heat sterilization you can search first post to find it quicker

Ok I guess I misread the tek, I see the wiki article now.(Dry heat sterilization) can this also be accomplished in a home oven? I washed my dishes really well then wrapped them in foil. Pc'ed them along with my bottle of agar. Then yes put them in my oven on a low heat for about a half hour. They were probably as wet when I took the out as when I put them in only now they were pretty hot. I still got less condensation then I did the first time I poured. Because I poured when the agar was around 119f. Instead of barely cool enough to hold.I'm doing my best to read, learn the search engine and not ask too many stupid questions. Thank you.

You don't need to do both you can either do dry heat or pressure cook/autoclave/wet heat
You can do dry heat with your oven yes. Its easier with a flow hood as it will cool them down and keep them sterile after you uncover the foil. They also come out dry
I don't like pressure cooking glass petris as there is no good way to dry them. If you bought an autoclave they are able to dry items and work really well for that purpose.

I will definitely try oven sterilization next time because it was a real pain dealing with the wet dishes. I'm kicking myself for not keeping a big autoclave I had in my 30s when I was learning to tattoo.I got it at a metal salvage for next to nothing and sold it for 50 bucks. It was a cool 50s avacado green one.

Hate to re-ignite this post, but it's the only spot I see anyone talking about the gl45 caps, with the 500ml bottle..I was just curious, can you refuse the caps and rings? If so,about how many times?

I have used the same caps and rings for quite sometime now. They definitely stand up to the pressure canner heat and I have had 0 issues to date. Media bottles are the way to go and last awhile.

3Rd transfers today.Washed, wrapped in foil and oven sterilized 30 dishes. 2 hours @ 350 to 375, while cooling I made my MEA in 2 jars. While they were cooling, I prepped my sab and showered, set my 2 jars in a water bath and checked periodically with my laser temp guage. Gloved up and unwrapped my dishes in sab. It was sooo nice to have dry plates to work with. When jars were at 120 I started pouring my 30 plates. Once they gelled I transferred from my 10 second transfers. I feel like I getting a little smoother. Still struggling a bit trying to get the sections off my xacto blade. My dish wrapping motion is improving. Can't thank you enough for the wealth of information you have provided here Bod.

How can I sterilize plastic petris. I just got ten small plastic petris. So I usually do the agar in baby food jars and use Patsy no pour. So I should PC agar in a jar and pour it into dishes. But can I just clean them with bleach or alcohol?

Plastic Petri dishes come pre sterilized, usually, in the bag. They are disposable. So essentially you get your Petri dishes, only open them in your SAB. Pour agar into dishes, keep them closed till needed. After use, you toss them. No way to reuse.
If you’ve already opened them and looked at them outside whatever bag they came sealed in, they’re dirty. I wouldn’t use them - there’s no way to sanitize them sufficiently now (pressure cooking or oven baking them will melt them).
If you want reusable Petri dishes you’ll need to get ones made of borosilicate glass (aka Pyrex)

Cleaning them with alcohol or bleach isn't good enough it will contaminate them. Those chemicals are sanitizers not sterilizers

Just wondering when I sterilize the agar for 20 mins do I still have to vent the PC for 10 mins before hand?
