Ideally you want to use your plates before they fully colonize the plate. The walls of the plate can harbor contams that remain unseen. And I'd recommend against putting your plates in the fridge immediately. You want it to grow a bit to ensure it's clean.

Thank you for the answer stareatclouds. Just seen this now. I’ll make sure to put this info to use from now on.
Another question... I have two bottles of agar in the fridge. If I want to heat one up for ten plates and put the other half back in, how many times can I heat the agar back up? Does it ever loose nutrients reheating it? Also how long can you keep unused agar in the fridge?

I apologize if this question has already been answered ad nauseum, but Im still having a little bit of trouble getting a real baseline recipe for agar. I have tried several different ratios, is 10g agar powder, 10g nutrient (LME powder or potato flakes or BRF) and 1L water the standard? Im confused because then bod says 20g agar to 1L grain water, which seems like it would be too thick? Thanks for any help!

just chiming in because you said you were confused about bod's answer and then only bod answered you. i could see how this might not help you be less confused. you should trust bodhi though. he knows his shit. i have done LOTS AND LOTS of agar and can also attest to 2% agar being legit. i use 2% always... unless i have some stubborn spores and want to make some softer agar. 2% agar 2% malt works like a charm for me.

May also depend on agar brand. I made 1% 'soft' agar the other day and couldn't even break up a huge wedge inside BLI jar after shaking violently for 30 seconds. I see other grkeers use 1.5% and tbeir wedges more or less melt when shook inskde jars. Last time I made 0.75% Agar and I look forward to using it as inoculate soon to see if it will break up easily or not. I think it will, I accidentally ripped holes in several plates while trying to streak a swab 2 days ago.
But yeah others have mentioned this as well, that agar brands can differ In 'quality'

so after I soak my grain I want to use the water from that and boil it then use it? or do I mix with agar and the pc it for 20mins or to 45mins

Thanks for your replies! I was pretty sure that this was the correct ratio but wanted to double check before I made any more plates. I will be using 10g LME powder and 10g Telephone brand agar powder in each of my 500ml bottles. If I use grain water I will just use 10g agar powder to 500 ml grain water because the grain water already has the nutrients.
I know c10 recommends using less nutrients, because the comparatively nutrient-poor environment will cause the myc grow faster in search of more food. This seems like a good idea in theory and something I would like to try out in the future once I can get consistent results with a 2%A-A/2%nutrient formula. Does anyone else have experience with more nutrient-poor plates? How can a system like this be put into place using grain water? Diluting? Thanks again!

Mine seems to be working fine and hamloaf's notes that I followed suggested diluting it as well. Have you bad experience? Or just had better experience going straight grain water with no dilution?

not all grain water is the same. obviously some grain water will be way more nutritious than other grain water. how much water, soaked for how long, how much grain.... i've cut grain water and not liked it at all. actually, i just don't like it at all even uncut. malt extract is SO CHEAP and SO NICE. any ways...
on the subject of lowering nutes to promote growth. i've fooled around with it a bit and am not too convinced. people say a lot of things but i get super rhizo fast growth on 2% MEA all day. i have noticed low nute plates just looking weak and sad though.

hey bod, i got the corning glass media bottles. two questions:
which was does the ring go?? and is the ring pc'able? they have a red version that is for high temp. Mine are clear.

The ring is so there's no drips. It stays on the bottle and gets PC'd same material as the cap. Polypropylene.
No ring and agar dribbles down the side of the bottle when you pour
