I purchased a similar bottle that is a little thinner and labeled as easy pour. They also run a buck cheaper. For us old folks this is easy to maneuver and pours cleanly.

Where do you order your 500ml media bottles? I’ve seen these with the blue caps and rings before but only on amazon and only 250ml I can’t find the 500ml anywhere.

Thanks! one more question, how long do spores take to "germinate"(?) in agar? This is the first time doing spore syringe to agar, I have always started with LC and then move to agar, the process is a bit faster IG.
How long does it take to spores to do something on agar? on average?

A 24 hours to a week or more.

yeah, it all depends how healthy and/or young the spores are. fresh healthy spores can show germination in a day or two. i've also had spores chill for a month and then germ so... somewhere between a day and a month.

Your attention to specificity is admirable. Relying on others for a microwave choice on the other hand..

If you can tilt it enough and watch it well enough for the flash-boil, you'll be fine. It gets messy quick once you get bubbles.

Sorry if this is a stupid question, but does it matter if your spores are dry or hydrated when you use the inoculation loop to put them on agar? I just did it for the first time like 5 minutes ago and what I did was sterilize water, put a few drops on my dry spore print, then swab the inoculation loop on it, then the zig zag on the agar. Reason I ask is because I noticed in your video, it looked like you were doing it with dry spores, but here you talked about putting them in the shot glass if from a syringe. So it shouldn't matter wet or dry right?
Also I was about to do another one with live tissue. I watched the let's grow mushrooms videos, and he tears the cap open, cuts up tissue, then sets it on top of the agar. Do I have that right?

If spores are old/dry, germination can be tougher, so more moisture may be needed. But I think making "softer" agar (less agar per volume of water) is better than getting your spore print wet.
For tissue samples, you tear open a fruit (as the inside hasn't been exposed to the outer air) and scrape a piece of tissue out and place it on agar, yes.

i would not recommend putting drips of water on your print. prints are not perfectly clean and water running around will just act as a contam superhighway (in my opinion). also, hydrating spores is not just a matter of getting them wet for a second. they need to "drink" the water and get it in them. this can take a little time. for this reason we either put old/dry spores on soft agar, like stare said, or scrape some off of the print into sterile water to let them hydrate for a while before putting to agar.

What recipe do you use for your brown rice flour agar? flour(g), agar(g), city water (ml)? Does the brand agar change things or are they fairly consistent? Thanks.

I’ve been using this exact recipe successfully for months. I prefer GWA simply bc it’s clearer and easier to see contams, but I don’t always have the grain water on hand.

Hey Bodhi 2 questions 
Switching from PDA to Oat Water for my Agar nute medium.
1. Will there be a noticeable difference between the growing mediums besides the coloring? anything I should know?
2. How much should I dilute the Oat Water? 50/50 Water/OatWater?
