I'm struggling with getting my agar right atm. I have a few plates i knocked on the 21st and they have started to colonize about a week ago but they are really really slow

Newb question here. I am having good success with my first spore to agar inoculation and subsequent transfers. I am meticulously wrapping each dish with parafilm. Does that stifle the necessary gas exchange for optimal colonization of the plate?

depends on what you mean by "meticulously wrapping". each wrap restricts more air. how much are you wrapping. one wrap is fine but some do two to be "safe". i wrap twice with glad cling wrap

Agar plates don't need gas exchange. I do 2. That's how bod does it in his dish video.

hey BOD....thanks for the tek, I'm going to give agar a try for the first time. I have all I need as far as materials but I have a question....what about the petri dishes....I have a pack of glass dishes but I don't see anyone explaining whether or not you need to sterilize the dishes first before you pour.....my heart and mind says yes but.......if so, what is a good procedure for this. Can it be done the same time you sterilize your agar.

i'm not bod but i know the answers. yes they need sterilized unless they are new, came sterile and are unopened. yes they can be sterilized along with you agar but you must be sure that they remain sterile. they can be wrapped in foil, sterilized and then unwrapped once inside the SAB or in the flow.

ahh, yes thank you. Never considered wrapping them up.......this tek calls for 20 min @ 15lbs to sterilize the agar.....that's going to be enough for the dishes as well?

yep. more than enough. you don't have to wrap them but you do have to do something to ensure that they don't become dirty after sterilizing them. contams are everywhere and wrapping them nicely in foil is a really effective and easy way to protect them.

I am going to be using my pressure cooker after sterilization to do the cool down procedure, so wrapping them up will keep them clean until everything goes in the SAB.....thanks for the heads up......ok, I'm going in......

Under glass dish sterilization on the first post in this thread. But yes they need to be sterile or they'll contaminate your agar

Good read. I'm doing a SGFC grow, and a shoebox grow as soon as my PF TEK jars finish colonizing, and i'd like to learn this so I can clone for the next one. So much to learn lol
