they might be and they might not be. pics would help but yes, there can be hidden (if you don't know what to look for) contams on a fully colonized plate.

Is it okay to run just one 500ml bottle in the pc or do you need to have it more full then that? Going to start my first plates soon.

I think so long as the bottle has something keeping it from falling over or something while in the PC it will be fine, that way water doesn't get into it if that is even possible with the container you use. I've yet to do any of this yet but from what I've read that seems like an answer that might help you someone could clarify though hopefully.

Yea the one I quoted under.. mine look identical to his but at 90% I don’t want to do my 3rd Transfer if looking like that is good enough to drop in jar .. thanks

I think that was showing a plate with hidden contaminates but I'm not sure. Sent him a message to point him this way for you. Since he hasn't had a chance to drop you a line refer to the link below.

First off thank you for the write up second if it's 20g/L brf That makes it 10g/ per bottle right?
And if there is sediment issues could you just put a little less or does it not really affect the agar ?

it's hard to say for sure if that plate is clean or has some contams hiding. i'd say it's clean but some would say that it looks suspicious. if you have plates that look like that you are probably good to go but to be sure, post a picture and let us have a look.

it's not my opinion but i know some that would say that the fuzzy undefined part surrounding the transfer could be indicative of some bacteria and some would also say that cultures that go extremely rhizo like that could also be running from bacteria.
again, i don't really think that but some people around here would. i'd say it's probably clean but i would take another transfer and watch it closely just to be sure.

Oh I wasn’t arguing, I just have a couple identical looking to his, and I’m newish to agar.. and was hoping mine was clean to, was gonna drop some chunks to grain and try making a slant.. but maybe I rushing it

if you are new to agar and have a couple plates looking just like that then you a rockin the fuck out man. good job. you can always drop most of it to grain and keep a little on a couple of plates.

Not really lol , I usually just multispore to grain works good in winter months but in summer trich issues are horrible. Only reason I’m doing agar is to get something clean.. lol everyone hates MS to grain but in winter months I have very good success not perfect but good.. I actually have a black mold in a spawn bag I’m gonna up load a pic I have never seen in my life looks like coffee ground patches ,, but never put an coffee grounds in with my grains. Lol
