Thanks! Yes they are from a spore syringe. I am waiting for my mini tupper ware pasty plates to dry out a little before attempting my first transfer later today.

Yeah as soon as you have time just take a small piece from each plate and let it grow for a few days on the next plate to see how it looks

Thanks so much for this TEK Bod. Here is a pic of a couple plates that I transferred on 6-25 off of my MS Plates. All are PE..



Cut these up and put them to grain jars today in the SAB. Stoked to see what they look like in a week!!

Edit. 3 hours later and transferring to new plates is not as easy as i was expecting! pretty certain 2 of the 6 plates have no transferred material and look like Freddy Kruger has had his hand in it. On a positive note poured these in the hope that i haven't totally f-up the original plate and the 6 new ones.

OK, managed a better photo of the other plate does this look like bacteria as i am concerned about the wispy star shaped bits! Cheers guys.

When you say 20g/L is that twenty grams to a liter of water? So it would be 10g for a 500ml media bottle? Just making sure I read it right. Also got myself a microwave and am going to get a mini fridge for storage, media bottle, and some dishes. I want to get serious with agar after trying pasty plates and having some success.

My agar iskills are definitely improving
1st pour:

Next 3 generations of pours, tried pasty plates, didn’t like them, moved back to peteris, changed recipes and added food coloring...

I love this shit!

looks cool. well made. was $17.90 so a bit pricey for a bottle.

Wow, not sure why I was afraid to start out with agar. Agar should be the only way to start this hobby. First agar plates were a success!

Thanks for all the info...I rarely reply because most of my questions are answered somewhere here. My problem is that every time I've tried to introduce mycelium grown on agar I get a contaminated jar. I use a SAB, bleached, a flame-sterilized blade, and I try to make sure everything is clean. Mostly I get a fuzzy white contamination, sometimes green but not the deep green that usually gets on things. I use both BRF and rye berries. Trying oats next. Any help is appreciated! Also, what is your favorite casing?

Bod, just wanted to say thank you for posting this (among many) comprehensive and easy to use TEK. I used this information to build a SAB, but the necessary ingredients to make some BRF agar (left over from my first forey into amateur mycology via PF-TEK), pour plates, and clone pins from a B+ MS BRF cake.
This weekend while out in the woods with my kids we stumbled upon some Phoenix Oysters growing out of a downed log.

Took three separate aseptic (hopefully!) tissue samples and put them to agar.

So, I assume by "dry ingredients" you mean just the agar-agar powder?
You say you put that in the bottle and then fill with room temperature tap water till 400ml and then sterilize.
But where do you add the nutrients? I am going to use my grain soak water, but you never mention when to add that? wouldn't you add that instead of tap water?
Pardon me if I am misunderstanding but it doesn't seem very clear.

Can I do this without a thermometer? I'm wondering if that's really that difficult or if you can just eyeball the agar/temperature.
Another thing, are plastic petri dishes fine, and, can I reuse them?
