I guess i'll try a small batch for a shorter time. 45m is the time others use with Mycomedia, it's pretty popular. and Fungi Perfecti quoted it at 45m as well. Who know's though (probably shroomery)

Nephrim said: Quick question for bod or anyone else. I get lots of boil over while my agar is pc'ing. I use 1L erlenmyer flask. I've tried it full, half full, etc. I always lose about 60%-80% agar by the time I remove it. I use Mycomedia from fungi perfecti which I believe to be a malt agar. It's recommended 50g powder to 1L water. setting, growing, etc are all perfect and I enjoy using it, just damn boil over is killing me. I mix, put tin foil over the lid. I've tried putting the flask in the pc while the water heats up and also placing it in pc just before I put the lid on and tighten, both boiled over. I PC at 15psi for 40-45m as recommended. I wait 1-2 hours to let the pc and agar cool together before even touching the pc. Then I open in front of my flow hood. The boil over happens early into the PC because it gets all over the inside of my pc and burns. Is the pc too hot? too long? I've tried with a silicone cork in the top of tubes I pc'd for slants and they popped the top within 10 minutes of the pc (I could hear them pop). I've filled water up to the same level of agar on inside of flask. Not sure what i'm missing. Thoughts?
Try bringing the heat up slower

It's about the cool down/depressurization How it comes up to pressure doesn't matter

I understand this, but then why does the agar explode from the beginning and burn in the pot? If it was just during depressurization it wouldn't get burned into the pot like it is. But who knows!
Did another round. 600ml, mixed agar with hot water til dissolved, filled pc to same or just slightly higher than agar inside jar. Tinfoil over lid with some wiggle room and a washcloth over the top of the jar to hold it down so it didn't float or bobble. PC'd for half the time I usually do, slightly over 20 minutes. No spilling this time. Still set fine and I'll see if there's a difference in contamination of these ones and the 45m ones. Done about 80 cultures the past few days, only 1 contamination so far. and 20 of those plates were from original contaminated plates that I was isolating from to save the genetics. and the 1 contaminated on was part of those.

I don't get why you think it's boiling over coming up to pressure just because its burned to the inside. The agar is hotter than the water as the water cools first touching the walls of the PC. This causes boil over. Hence using 3q of water in a 23q PC.
Chances are you're not using nearly enough water in the PC or trying to do too big a volume of agar in too small a volume PC.

I believe you're right, I didn't have enough water or too much agar mixed with too long of PC times. This last time went smoothly with none lost.
I thought it exploded on the come up because the walls of my PC we're blackened from agar cooking on them that had spilled out and also because I could here the corks pop on my tubes in the first few minutes. But none of that matters, point is it's fixed now and you know best. Thanks for the help as always!

Hey bod, where do you like to get your petri's in bulk from? I've been searching the past few days but mostly find 500 for 100-150$
Edit: I feel like a jackass, but they showed up after several pages online.

Step 2 - Let Dry
Step 3 - Place them inside your SAB

there is no real way to reuse plastic petris, u cant PC em or they melt, therefore 'sterilizing them' is out of the question they are not made from pp5 and the ones that are are expensive.
just letting u know from what ive gathered around the site. even if u replaced the dawn soap with ISO it still wouldn't be considered sterile.

I don't know if you have actually tried that, but I have. Congrats if that worked for you but I'd say you'll probably grow a lot of NOT mushrooms. I rinsed thoroughly with iso in front of a hood, let dry, poured, inoculated with nothing, wrapped, and grew mold.
