how to inoculate agar dishes

Hello... Quick question...I followed pasty's agar tek, and have the pda jars cooling right now in the pc...my question is, I didn't c what the best way to inoculate the plates would b?

  I have prints, and one homemade spore syringe. Can I just simply scrape a few spores in there and close it up?

Yup. It's best done ina GB or SAB. But you can use the syringe or a little scraping of a print. I do believe the syringe is the better way to go tho, you spread less spores out which is what you want.

A drop from a spore syringe will spread around on the agar and it can kind of make it hard to pick out clean/healthy growth sometimes.

If you use the syringe squirt a drop on a loop or a sterile cotton swab first.

To inoculate from the print, flame your loop, cool in a blank plate and swipe the print.. the agar will help pick up spores... then swipe the reciving plate.
Swiping is good, youll have an idea of where to expect germination

Means after flaming the loop, plunge the hot metal into a section of agar off to the side of the plate. This cools the tool so you can use it without damaging the spores. It also leaves melted agar on the loop, so the spores stick to the agar coating it.


Buy RR's Let's Grow Mushrooms videos. Very good investment to make, I still watch them from time to time.

Yes... the videos are definitely a good investment. They are a great reference tool. Though I have been into the hobby for a little while, I watch a vid(s) to freshen up or look up some info quite often.

Alright, so I made the pda mixture, pc'd, then let cool overnight...when I got them out, the bottom half solidified, top half was still liquid...what gives..I measured out the formula Damn near to the mg on my digital scale.  Cooked on low. Added food color, also I can see that any sediment (I assume potato flakes that didn't fully dissolve) where all contained in the solid(bottom) section. I put them in fridge, and they are solidified all the way though, are they still good to use? Even tho I can c that all the sediment is in the bottom half, top half completely clear, besides food coloring.

Ty, I understand there is allot of questions, and I really appreciate the help!!!! :-)

Sediment is fine. Mine look that way when I use instant potato flakes.

You're trying to isolate mycelium I take it? You want to do what's called a streak plate. The basic idea is to spread everything out on the surface of the agar so individual colony forming organisms are far apart before they start growing. That way when stuff has grown for a bit it won't all be jumbled together, and you can cut the sections you want out of the agar. Here's a video for the 4 streak method:




You probably don't need to do 4 streaks (2 or three might work fine). Make sure you re-sterilize your loop after each streak.

IME pastys agar tek gave me realm watery agar. I still use that tek to this day but I use double the agar he calls for. I don't have sediment and I don't have lots of condensation.

If you put a folded piece of paper towel between the lid and foil it helps alleviate condensation and/or steam getting into the container.

20g/L of agar-agar should give you appropriate consitency. 15g/L is most lab grade. if you buy agar from the store make sure it doesn't have added ingredients or sugar otherwise you're not adding as much as you think you are.

Thank u guys so much for all the speedy answers...Ok..so feel like the more I learn, the more questions I think of..lol. but I have a pe swab...do I just streak it the same way I would if using an inoculation loop?  Also, I've only worked with grain in all of my past grows... Recently I had a bunch of brf/verm given to me...I was obviously familiar with pf-tek...but never tried it...when I went to look up ratios of Verm/brf/h2o....I read a few different answers...so, what ratio do u guys use?  I really want to try and "perfect"every technique...and recently ran into some extra funds so I can buy whatever is needed.  Also, how long do ur guys brf jars take to show initial growth? I'm on day 4/5 with no noticeable growth...starting to get worried..sorry for the long one paragraph form..lol. 2mg clonazolam, &2mg alprazolam, and a couple beers..my grammar/writing skills go right out the window...lol

I believe the streak is the same but wait for one of them others to clear that up. I use 1 part water 1 part flour mix that really good and add 2 parts verm I also add in a pinch or two of gypsum but there are a few different additives. I see growth in about a week so id say your fine still.

Use your own prints. Read the tiger drop tek in my signature.  If you use pre-sterilized cotton swabs you won't even need to flame.  If the print was taken with any sort of care you should be able to get clean growth on your initial inoculation and then you can drop the whole agar plate into a grain jar.
