Birdseed recovery problem...
    #2191097 - 12/19/03 06:53 PM (12 years, 11 months ago)

Recently all my cloned WBS jars have not been recovering properly from shaking. I then did a test of a fully colonized cloned KST3 jar (WBS) and found that it did not recover at all, in fact, there seems to be absolutely no growth at all, I did not shake them to vigorously nor is there the problem of contamination, it seems that after I shake the jars it becomes a little wetter (almost gooey) inside, and it kinda looks like a water contam, but I am doubtful of this as the jar was once a fully colonized jar.
Any suggestion/comments appreciated.

Thanks
TeKn0

This has happened to me; invariably a smell develops. . .bacterial contamination. IME, it can be smelled though a filter disk, though it may take a few days to build up.

What are you using for filters, are you using 2/3 cup of birdseed to 1/3 cup water, and how long and at what pressure are you sterilizing your grain? Are you using agar or are you putting spores in the jars? Are you using a flowhood? There is something in your lab technique that is causing a bacterial contam. Are you showering directly before you you do all lab work. Hell, it does'nt hurt to gargle with Listerine.

goo and poor recovery is a sure sign of either bacterial or yeast contam,it happened to me on my first trys with grain.there is only one solution......get a different brand/type of grain, birdseed, millet etc.once i switched brands,i've had no problems and all my prep methods were identicle in each case.

Alright thanx for the replies.
Just to answer a few questions, I was using agar to spawn the grain. I use H202, but don't like bleach, and I have been using grain for quite a while now with no problems.
I think hanky hit it when he said the brand of WBS I chose could be the culprit, as I had just changed brands, so I'm gonna try another type of spawn for the time being maybe millet or another brand of WBS.
Thanks.
