cloning question

How long would a tissue take to colonize the jar?

Depends on how large of a tissue sample and how well you prepped the grain jar. But I would definitely clone the mushroom to an agar plate first to be sure there aren't any contaminates, wait for it to colonize, then transfer a healthy piece to the grain jar.

Could it happen that sometimes the tissue wouldnt colonize?

If you take the tissue from inside the stem of a freshly picked mushroom, I don't see why it wouldn't colonize. The mycelium is still active and putting it on a nutritious medium, such as an agar plate, would trigger more growth. Try it out and post your results!!

On the rare occasion I have a set of jars not colonize it's because there was a hidden contaminant on the plate I took wedges from. What is your cloning process/sterile technique like?

Well iam cloning to a sterilized half pint jar of grains (cant work with agar now) i sterilized the knife in a pc wearing gloves sanitised with alchol and transfered the tissue inside a sab.
