Is my liquid culture looking good ?
    #21895818 - 07/04/15 07:36 AM (8 years, 6 months ago)

Hi !

I've just started my first liquid culture and was wondering if it is looking good ? I have used 1 round tablespoon of extra light malt extract to 1 cup of water. I made my water boil 5 minutes to "pre-sterilize" it, then dropped 1 cup of water in my jar and added the malt extract. I did wait a little for the water to cool down a bit then closed my jar loosely and sterilized in pc 20 minutes at 15 psi. Once fully cooled I injected 1 cc of spores.

It look tick to me ? And is it normal to have all those deposit in the bottom ? (I have a magnetic stirrer thing in it too)

In front of a light :

Thanks for helping

The deposits at the bottom are just sediment, and it's cool.

The facts that you've inoculated your LC with spores, coupled with the way the media can't be seen through are bothersome.

How long ago did you inoculate the media?

Just minutes ago. I was only wondering if it look like a viable liquid culture or if it is too dark or something like this

Anyway, if the sediment is ok, I guess i'll just wait and see if it goes well in time. Thanks !

It is probably a viable media but the concern is that it was MS inoculated. Syringes from spore prints almost always will have some nasty contams in them which often multiply in a liquid culture much faster than mycelium. Ideally you would start with a clean wedge from agar.

:whathesaid: then tested for cleanliness on a few agar plates. That's what you do for LC. I wouldn't get your hopes up for that one. Almost every syringe plate turns bacterial.
