If he can't get used to using a SAB he's never going to be able to do g2gs or use agar.

Still air boxes aren't exactly hard to use, either, which makes me question if his inoculant is to blame and not his sterile procedures.

With statements like this, he's a prime candidate for benefit from a SAB and some agar(to clean up dirty spores):

Op, learn agar. Acquire a clean culture, people here can help you do that. Once you have a clean culture, prep your own grains and inoculate in a SAB. You'll get success, it's not rocket science.

A thought occurs. What if he has a leak in his SAB? If there was a leak by the gloves, every time he moved he'd be pulling air in.

OP could you post pictures of your SAB? Just spitballing here I would sprinkle some baby powder on the outside around the seals for the gloves, then get in there and start moving them around. Then check for powder making it inside. Just an idea

You shouldn't use gloves when you work with a still air box, for a few reasons.

1) You won't be able to remove your utensils to the outside of your still air box to flame sterilize them. Common tools that need to be flame sterilized during SAB procedures are scalpels, syringe needles, and inoculation loops.

2) You may end up creating a sucking effect as you move your arms around, causing air to suck in through the lid from the outside. Nobody even makes there still air boxes "air tight" that I know of, how could you feasibly? Still air boxes don't need to even be air tight to work, they work because the air is still while you work. I've done dozens and dozens of g2g transfers with a 99 percent success rate. I have no attached glovesl, my SAB isn't air tight.

The best way to use a still air box is to spray the bottom and sides down with soapy water(I just use plain water) or bleach water. The wet walls helps contaminants stick to the bottom/sides so they're not floating around as you move and work. Load up your SAB and then wait 20 minutes, let the air settle. Go to work carefully, but swiftly. Anything opened should be opened for as minimal a time as possible, and never place your fingers/hands/arms over anything you want to remain sterile(open jars, petris, tools,etc).

Interesting. Perhaps this is why I failed. I was looking at your gallery, did you just put in the sewer flanges with nothing attached?

So you just reach in to do your work while the box just protects the ambient air from contamination? I would think reaching in and out of the box could spread as many contaminates as open air... Its been a long time since I've been in the hobby. Back then you would see plastic bags or dishwasher gloves on the inside. I love the ever evolving knowledge we get here =)

I don't attach anything to the flanges. I like the flanges for a few reasons:

1. They are comfortable. Sharp edges on a tote hole aren't fun to rest on while you work.

2. I feel they do a bit to keep outside air out as you work, as they extend inward and outward a bit.

3. I screw them shut with the accompanying cover when not in use. Helps keep it from getting too dusty inside. I also close it up for the 20 minutes while air settles, though that's probably overkill.

Have you ever worked with agar? Have you ever done a g2g? Do you do them successfully in open air?

Nah, I never got that far. My status is strictly amateur. Thats why I like asking questions and having discussions. Learned quite a bit from you tonight, cheers man. I'm gonna have to look at building a box at some point now =D

Op, are you using a dust mask? Try using one or simply use a bandanna around your mouth. Sometimes your breathing will push in contams into your SAB.
