Photo of bacteria on plates
#21865514 - 06/27/15 06:45 PM (8 years, 7 months ago) |

2 of the plates were from a sloppy pour I guess, never poured a bad plate before
Anyways a lot of folks ask me what bacteria look like on agar. So I figured I start a thread where people can put up photos of bacteria in plates and jars, mold colonies etc
What to look for

Trich never looks like that on plates in my experience. Also rarely does it turn green on a plate if it's growing with mycelium too.

Trich usually has a crystaline texture to the surface and a bubbling growth pattern. Also like bodhi said it usually needs some decent FAE to sporulate.

On substrates though powder green look can indeed be a multitude of species and even a few different families. Aspergillus, penicillium, trichoderma.. They all can range in different shades too. The online mushroom community generally calls them all trich. Realistically speaking a microscope would for sure tell what it is.

All three of the aforementioned contam species have their own shade of green. Been blessed to not run into too much Aspergillus, but have seen a fair share of penicillum and trichoderma. Penicillum is a teal green, where trichoderma is a dark, deep, forest green color. Very distinct. Trichoderma is the bane if the mushroom culture. HATE!

Interesting. I've also seen a bright green/mossy green look in certain species of trich. However I know what you mean by the teal look in penicillium. I've seen that quite a bit too.
So looking at bods pic, I can see it's a very fine powdery look. Almost looks like a smooth surface, whereas trich has more visible particle/granite look. Is this a key distinction between the 2 on substrates? Also another distinction as mentioned before is that it doesn't sporulate on plates well. Just curious how you guys visually tell the difference. *runs off to look at pics of them*

I don't know what the guys are talking about over the issue of trich not sporulating on agar very often, because I'v had trich break out on several plates over the last 6 years. Recently had a contam breakout in the lab. 40 plus pre-poured plates that were sitting for over a month and a half ALL had trich in them. Don't know how long said trich was growing on the plates for. What is known is as soon as the contam was discovered it was immediately discarded. Black mold is another that'll sporulate all over your agar media.

A few months ago I was running into what I think was Aspergillus, it is a nasty one too. A little darker green than trich and it seemed to pretty much start off on the very surface as little green speckles.. instead of white like trich. It would turn myc yellow brown wherever it touched it. Not as nasty as trich but certainly ruined a few tubs. It still turns up every couple grows too

A question, Bodhista. The plate on the right... Is the mycelium also contaminated? I have a few plates, on their 3rd and 4th transfers away from clones, with no glaringly obvious contams, with big differentials like that...

I should have saved my plate from earlier today and taken a pic. It had a nasty chunk of bacteria on it. Thank god I've gotten into agar. It's so nice knowing that I can just transfer healthy mycelium away onto another plate and not worry about it.

It's the very light film covering the "uncolonized" area. I don't know how many dishes like that slipped by before I finally noticed it but I started to wonder why the mycelium was suddenly stalling instead of colonizing the whole plate

When ur transferring out from the contaminated plate to a new one, do u cut all of the good myc out for the transfer? Or just a wedge. I'm under the impression the goal here is to have a plate of full myc, so I would think you'd cut all of it....haven't dine agar yet, 2 or 3 more days til clones are ready, and just wanna be sure I'm at least starting it right.

Just a timy wedge dude. Like a grain of rice. This way you can cut away from contams, isolate genetics etc..
It will grow out on the new plat

Maybe this will help a little more, if you look at the quart jar here, if you notice you'll see its got that thick, creamy cottage cheese type look that comes from bacterial infection. Oddly enough this jar did not smell off at all.
I only have this one picture of clean spawn (gallon jars) to compare to right now, and its still not the best photo that was my first go with gallons and the GE hole in the lid was too small. But you can see the difference yes?

Also in those first two jars the spikes of mycelium spread throughout are a sign of bacteria... Myc can look rhizomorphic but it will still remain individual strands when you look at it closely... Those right there are all fused into single chunks... The plates that have those random tufts of myc that are raised up from the rest are also signs of bacteria as far as I can figure.

See how thick and white the myc looks? Sometimes it even gets a cottage cheese look. It will look solid like a block sometimes too. This photo below is another good example

This is the only photo of clean spawn I have at the sec but you can see the difference.

Those thick white spikes of mycelium are a give away... In clean spawn the myc will be a bunch of individual stands when you look close... Those look like they have hair gel on them...

I think I've seen some of these. What are their names? Especially the dark green one in the upper right corner.

2 of the plates were from a sloppy pour I guess, never poured a bad plate before
Anyways a lot of folks ask me what bacteria look like on agar. So I figured I start a thread where people can put up photos of bacteria in plates and jars, mold colonies etc
What to look for

your pictures are broken.
