Isolating question

I'm planning on moving away from MS and onto agar. First I am wanting to isolate. I have pretty much the teks I am going to follow, but I have some questions. When you start sectoring out and transferring to different plates when do you start fruiting? Do you wait until you get a pure isolate to start? And what do you do with all the plates that you took wedges from that weren't isolates? Just toss them?
I'm planning on starting with a cloned fruit from my current BRF cakes. And isolate from there. I've heard it's easier then starting from MS directly.

1) whats the point of isolation if you dont isolate.
so yes, you wait until you have a single growth covering the whole plate.
2) theres many ways you can do it, you can make a live culture/live inoculate from the plate... or just drop a wedge onto the grains.
yes you can toss the partial plates, you dont need them.
but you CAN use them to isolate OTHER strains and compare good characteristics (like fast growth) and use the ones with traits you like.

Thanks you are awesome. Is it harder to isolate from a MS syringe then from a cloned fruit? In my mind it would seem to be because how would you know if you have fruiting mycelium or not.

Non fruiting is rare. Clones may be closer to isolates or you may get lucky and a fruit Clone may be an isolated first shot. It's best to just fruit clones. Isolate in your spare time most isolates suck for every 100 isolates maybe a few are worth keeping.

I gave up finding isolates. I used to always get better results with clones as the genetics seemed complimentary

Im with Bodhisatta about making strain isolation, more of a second thought.
When I start with a new print, I put to agar to get clean spawn and do MS grows to search out awesome clones... I do further work with those clones and usually do my grows with those.
But at the same time I will keep transfering from the original culture from spores and isolate.
Once youve made enough transfers that you see obvious sectoring, isolate out a piece of each sector to a blank plate. If your culture no longer shows *any* visible sectoring than youve got an isolate, you will want to put this aside to test. If it any of the plates still show sectoring you will want to isolate further might take another transfer or two
I try to get 8 to 10 isolates out of a culture to test out. Not all gunna rock out.. Ive been saving the best one out of these to slant and then I start again from spore and do it again. Eventually youll find something awesome but as Im finding out it take a lot of work and dedicatiom but hey its fun
Its definitly worth it to isolate IMO. But for our purposes if you stabalize a rocking clone you can get the same consistent results
