Black poplar pinning in vitro
    #21849159 - 06/24/15 06:54 AM (8 years, 7 months ago)

Hi all,

So I got this black poplar culture a little while back and put a wedge on popcorn on 6/3 (I was out of rye).  This morning I noticed it's pinning...a lot!  What should I do?  I guess I could open the jar and try to get some fruits in vitro?  I was also thinking of opening it and transferring a few grains to rye with flame-sterilized tweezers.  Thoughts?

So what if it is pinning. There is no reason you couldn't spawn grains to substrate. But you could also fruit it straight from grains. But in this case you would get smaller yield. You would have to fruit it in environment with proper humidity, if grains dry out they can contaminate.

You now wanna G2G it? Sterilize tweezers in oven or pressure cooker, you can't sterilize whole tweezers just with flame. I mean you could but it's not that easy, you would be risking contams with new jar.

I was thinking about casing the jar maybe?  I don't know.  I have plenty of plates of this, so it wouldnt be terrible to get a smaller yeild from this.

I shouldn't really be surprised, this culture has pinned from every agar plate I've put it on as well.

You could restrict light to the sides/bottom and case the top of the jar.
