Agar Pour Tek

I've gotten almost all the answers I need before I feel confident in diving into agar and making my own plates.
However, I can't seem to find a good "pour" tek when it comes to mixing up agar. I'm dead set on using petri dishes as they are already in the mail.
From what I have pieced together from UTSE and reading from those results is this method. So! Will this work???:
1. Mix appropriate amount of LME, agar, water in a small pot with hot water until all is dissolved.
2. Pour into a glass bottle (Crown Royal in my case because it will fit in my PC)
3. Cover the top of the bottle with Tyvek and then foil.
4. PC (20min? maybe more than that?)
5. Let cool
6. Pour plates from bottle inside an SAB
Is this basically the idea? Am I missing anything?? I can't find any good bottle pouring teks, but RR mentions using a Crown Royal bottle and pouring from that.

doesn't have to be glass. I use a pp5 shaker bottle that one would make protein shakes in... you can find them at any health store or even supermarket. poked a hole in the snap on lid with a hot nail and siliconed a cut-out sfd on there.

3. Cover the top of the bottle with Tyvek and then foil.
if you use sfd, you dont need tyvek. top with foil

4. PC (20min? maybe more than that?)
what psi?

5. Let cool
cool enough to where you can hold the bottle comfortably, but not cool enough that it starts gelling
Quote:
6. Pour plates from bottle inside an SAB
give it a quick and smooth swirl and pour away. dont put your hand over the plates when they are open.

that is basically it. I like the shaker bottles because they have measurement lines in ml and the lid has a pour spout. good luck!

Been awhile since i used agar, but if i remember correctly.... taking a coffee mug of boiling water an putting it on top of a stack of cooling agar will stop condensation from forming on the lids.

I pc for 45 min. Wait about 2 hours after pc cycle is done to pour to reduce condensation on petri lids

OK good info. I'll PC at 15psi for 45min.
Also I'll pour when it's still hot, I did not know it would gel up that easily. Is there a temperature to aim for? 160°F+ ? ..since 140°F - 160°F is pasteurization temps? Or is this ONLY because agar gels up and becomes unpourable once cool?
Sksoul, if I used a Crown Royal bottle the only opening would be the top and I wouldn't be using an SFD. I would need some form of a lid that allows pressure change so that's why I figured take a piece of Tyvek and rubber band it around the bottle neck, then cover will foil to keep water droplets from raining down off the inside lid of the PC.
But I like you idea better of using a pp5 water bottle with measurements and drilling my own hole to make an SFD hole for pressure change.
How long to I let the poured plates cool before I can knock them up?

2 hrs? Are you leaving the bottle in the PC or something? I feel like my bottlewould start to gel up by then.
I take my bottle out once the pressure falls and I pour as soon as I can hold the bottle
I dont really get condensation problems, only the top plate or two. If I set a jar of hot water on top it resolves that but I dont usually bother

2 hours is a good round number. I don't really check the time like azur suggested, I pour when I can hold my pc from the sides with bare hands and not get burned.

you can poke a small hole in the crown royal cap as well. either stuff it super tight with polyfil, or use 4 or 5 layers of micropore tape. I don't think a rubber band around tyvek would hold tight enough for sterility, but admittedly, I've never used tyvek as a filter.

Yes. I leave everything (agar, grain, tools) in pc until it's cool enough to use. it's the most sterile place for them to be

Ah ok yea I gotcha now. I carry my PC upstairs and put my agar bottle in front of the hood untill I can touch it. It cools pretty quick

Stacking your plates after you pour them really helps cut down on condensation. Condensation is only limited to the top few plates, and if you wait long enough to pour you can end up with virtually no condensation.
Also, don't use tyvek or make a fancy lid for your bottle. Just put a cotton ball or some poly in the neck and then cover it with foil. Work quickly and methodically when pouring your plates. If you're using a SAB that you have set up all the time, you can keep the blank plates in there without wrapping them for quite some time. If you're gonna be storing your plates out in the open, I recommend wrapping them.

I could leave the plates in the SAB if I wanted. When I made it I fitted the hole with female PVC pipe fittings.. I can cap off the whole SAB. But I should I wrap them at some point regardless??

I poured my last plates at 130 F (used an infra red thermometer), which is the only temperature that I found, through extensive searching. Poured 44 plates. Last few ml gelled in the flask...
Straight away into ziplock sandwich bags. Plates sit for a few days, at least. Parafilm after inoculation.

This is my exact SAB lol, right down to the PVC clean-outs! In this case, you only need to wrap your plates once they're inoculated and you plan to remove them from the SAB.

This is the way I do it too, only I usually leave the freshly poured plates in front of the flowhood until the next day before inoculating. To each their own though. Whatever works. There is more than one way to skin a cat.

Well I don't have a flow hood so for now the plan is to leave them in the SAB until inoculation.
I don't have any parafilm but I do have GLAD Cling Wrap. I had my finger on the purchase button for the parafilm but when I saw it was $25 a roll I decided against it. Maybe when I have more money I'll order some.
Now if these petris could get here already! I'm getting anxious..

I prefer saran wrap to parafilm anyways. You'll be fine

Not to diss Saran Wrap, as I haven't used it, but that 250' roll of Parafilm goes a long way. Less than 6" gives me 1 1/2 wraps around a standard plastic petri...

I've read RR say that when half a plate was wrapped in parafilm and the other half in saran wrap, the parafilm kept the myc growth at a rapid pace the entire time while the saran wrap/(polysomethinglene) showed rapid growth only during the beginning, and slowed as it grew larger.
So that motivates me enough to want to try it at some point for sure.

That's the roll I was looking at. Lol

I have been using the same roll of saran wrap since I started in 09'.
I usually don't want or need my plates to grow rapidly.
The difference is minimal anyways. Talking a day or two
