Poor Colonization
    #21813948 - 06/16/15 09:45 AM (9 years, 7 months ago)

Alright, so I know I'm probably going to have a ton of people jump on me but I'd rather get answers than have my feelings hurt.

tldr; colonized 4 rye grain jars. myc growth is super slow... I colonized back on May 6th and only one of the jars is anywhere close to full colonization but it still looks dull and unhealthy. I've kept them at room temp (ranges from 78 - 85 most of the time) right in the middle of a bedroom with ambient light. What's going on here? I can post pics of what the jars look like later when I get home.

how did you sterilize? how did you inoculate: ms, lc, agar? did you use a sab, hood, or open air? try to post some pictures. sounds like you have a contam, most likely bacterial or you'd know by the off-colors.

Inoculated using spore syringe. Inoculated into pre-bought sterilzed rye grain jars and just kind of left them. I haven't opened them yet and don't see any discoloration so I didn't think it was a contam. Would it be poor growing conditions? The jars themselves always looked a little dry. The FAE is built into the jars I bought, .22um. Below is link to the jars. I'll be posting pics later today.

grain should look dry (they hold moisture within), shouldn't be a problem. using spore syringes is a crap shoot itself, let alone into grain jars you didn't sterilize yourself. nothing more you can do but wait, but after 5 weeks i'd call it a lost cause.

85 is really hot for incubation. id try to keep my jars at 75 max. you probably have some serious bacterial shit going on.

What's your preferred choice? Prints? Agar?

Also, how do you keep your jars at a decent temperature? I live in the south and right now it's getting harder and harder to keep it cool. Do you have an incubation chamber?

spores to agar, clean it up and on to grain. I prefer LI or LC made from clean agar, but some swear against LC.

I live in zone 5 so my high temps are around 85 outside, 75 inside at the moment. Fans and air conditioning help.

Dude I bought one of those back before I started making rye my self. I noticed them gas exchange filters don't really work and act like they are clogged. Make your own jars, just buy the rye on the internet and slap them in jars and drill a hole on the lid and cover with micropore tape.

That's what I was thinking, poor FAE. After five weeks do you think the rye is salvageable? Could I remove the lids and continue with successful colonization?

You could transfer to a clean jar with good gas exchange to let it further colonize. You could salvage if its dry enough, although I've never had any luck with salvaging old rye...

Tighten the lids then loosen them 1/4 of a turn...that is plenty of gas exchange for grain jars.

Here are the pics of the jars. The lighting is a bit off. The myc is a little more white and spread out than this but still half of it only looks like frosted cereal. Opened them up to air them out and they both smell pretty ok. Doesn't seem like a bacterial contam. I thinking of taking one of the two, pouring it into a new jar and mixing together with coir/coffee grinds to see if I can transfer to a bulk substrate. I don't mind do a little experimentation.

Perfect! That was it. I took your advice and loosened up the lids on those jars. Let them air out for a couple seconds and then only slightly put the lids back on. Over the past 4 - 5 days I've had tremendous mycelium growth. More than I've ever had for the past 30 days. It was definitely due to bad FAE. Below are pics of the jars now.
