Once you transfer from agar, do you toss it?
    #21840941 - 06/22/15 08:40 AM (8 years, 7 months ago)

Just wondering what everyone else does when doing agar transfers.  Typically I rewrap and keep the plate I transferred from, sometimes taking other transfers in the future.  Is this standard practice?  I know every time you open a plate you expose it to contams, so should I be transferring and tossing them?

I keep it around and check on it to see if any contamination grows. I'm very new to agar as well so please wait for the more experienced.

Get the sectors you want and toss the rest. If you feel the need to keep anything you do it on a new plate or a slant.

I keep everything, except the contamed ones. I use my old plates (sealed with cling wrap) as backup. And for cakes to grow on top of in my shotgun chamber. I think I have a hoarding problem.

Well, in my country we are allowed to have it all, except for the shrooms themselves.
And since I'm a beginner/noob in myco-stuff, I tend to make sure I have backups.
I just like to see what happens to slight contaminations, differences in media and their contents and what happens when plates are overrun by mycelium. In another room I have a plate of B+ that is preying on trichoderma. Since that's pretty unique as far as I could find, I decided to monitor it daily. I inoculated it with lipstick mold to see if it could fight that off as well. It seems to be losing the battle. But that same mycelium survived 45 mins in the PC (i was experimenting with re-sterilising jars). Now it seems invincible; a completely infected brown PDA plate is now being overrun by B+. Heck, I just don't ever want to toss it.

I have a tupperware box with old plates, and one with newer ones until I'm sure I don't need the old ones anymore. But it's just too damn imteresting to watch!

I'm pretty new to agar, so things may change... But, I keep mine, mostly to observe for contams, to monitor my sterile technique...
Sitting on 100 plates now, lol!

I did just the same thing... I once let a contaminated plate of trichoderma fruit. Let me tell you, that was not a fucking good idea. Long story short my whole bathroom ceiling got contaminated with trichoderma and I had to call the real estate agent to deal with it. Luckily there was evidently an air circulation problem, so they installed an extractor fan and repainted the roof. But seriously the roof was completely covered with thick trichoderma and I never did a grow in that apartment again.

If you see a contam plate just dispose of it carefully, don't let it grow out... and definitely don't let it fruit.

I guess the heart of my question is do you guys make repeated transfers from the same plate?  I have a few I've taken a wedge off of, but typically I keep them and do it again later.

If you save a plate, every time you open that plate your risk multiplies exponentially of having mold land in that plate.

And although it may not germinate on top of the mycelium where it landed, once you drop that piece in grain it will

I don't keep anything contaminated. Any potential contam is disposed of properly.

That single petri wasn't what fucked up your roof. It was likely the ceiling that contaminated your plate, not the other way around.

Possibly... that seems more likely now that you mention it lol! I was pretty stupid the way I handled the contamination though
