Adding gentamicin sulfate to spore solution
    #21831803 - 06/20/15 09:36 AM (8 years, 7 months ago)

Hey, I've got a simple question for anyone here who might know the answer.

I have a number of cubensis prints that are contaminated with bacteria.

I want to make a batch of spore solution out of them, but I need the bacteria to be nullified.

Would adding some gentamicin sulfate to the solution prior to autoclaving be an easy fix?

How much should Gentamicin should I add per liter of water?

Will it have an effect on spore germination?

I also have some Chloramphenicol.

I appreciate any input. Thank you.

I'm pretty sure the normal working concentration for gentamicin is 10ug/mL, maybe as high as 25 in some cases. That is 10 mg of gentamicin per liter of solution.

That said, gent' is really most effective against gram negatives. If your contamination is a gram positive you may not see spectacular results.

Chloramphenicol is more broad spectrum, though its less heat stable and you may lose a bit during sterilization (its not terribly fragile, but you could expect to lose ~15% or so)

Typical working concentrations of chloramphenicol are in the 12-25ug/mL range, maybe as high as 35ug in some cases. However it should be noted that it is bacteriostatic, not bacteriocidal, and upon dilution into your final substrate any bacteria present may "wake up". Or the fungus could eat them. I honestly have no idea what the outcome would be.

Hmm, I see. Thank you for the information. I'm going to add a bit of both to my solution and see what happens.

That shouldn't be a problem, mixed antibiotics are a fairly standard solution to tricky problems. Hopefully it works out for you.
