Anyone work with plant tissue culture?

Has anyone here worked with any plant tissue culture before? I'm curious on various protocols on cacti and other plant species.

Many of those posts are quite old. One user I messaged does other things now. So, I figured that I'll start a new thread for anyone to chime in.

I've attempted home micropropagation with limited success. I can usually get a sterile plant and callous but growth stops even after I transfer. I think I had too high levels of hormones.
For specific protocols you should just google 'species + micropropagation'; if you can't find a published article with the exact plant you are working with, just search a related plant and work from there.

That's what I've been doing, and I've found some really good ones. It would be great to have a Salvia D protocol, but one old user Una, won't give up his protocol =) I'll just do a full panel of various different hormones and concentrations to figure it out... it won't take too long.
I'll figure out as many protocols for various plant species as I can, and share them. That way we can have more people getting into the micropropagation hobby, and spread the love across the world. If anyone wants to help with that, post up =)

I'm down to get into this. I've thought about it with my cacti but I just haven't really looked into it much farther than the thought.
If I can see a success then I'm sure I'll hop in as well.

i look at plant material every day under a microscope (and a bunch of other shit bacteria, yeast, cells, etc) ... most times i am looking for parasites (fecal samples) and I have to determine what is plant/root material and debris. sometimes it will resemble a parasite. parasites will usually have a membrane around the outside though, so they're pretty easy to distinguish. I want a microscope at home so I can look at spores but I guess I can just start bringing them to work with me.

Have a look at these peoples site. They sell tissue cultured cacti and succulents. They are out of the Netherlands.. of course =) : http://www.succulent-tissue-culture.com/EN
So can you see the success they have had?? It's definitely a good industry to be in.

That's a very interesting, and important job you have there. People, or animals (not sure if you're looking at human or animal fecal samples) depend on your analysis of the given material, to formulate a conclusion as to what they are infected with, if anything at all. Better not be caught sleeping on the job there =)

I did some tissue cultures a couple years ago trying to clone cannabis from leaf and stem cuttings. I tried several batches and was careful to follow the instructions carefully but all that ever happened was mold took over the tissue sample. I haven't had enough time recently to keep at it.
If your going to try, be prepared for some possible trial and error work before you get it right.

Thank you for the advice. I'm sorry to hear about your troubles. Were you working under a flowhood? Did you use PPM in your media to assist with contams? Did you rethink every step to see where in your technique needed improvement, to overcome that issue?

I used PPM yea, but just a SAB, not a flow hood. My SAB abilities were never really that great, I always hated how restrictive they were and I tended to get impatient with the whole process. I guess you have to be very careful/patient to get it to work.
