Need your help w/ Amazonian

Hey Guys 'n Gals,
Lately I have been trying to find my way around in this hobby (or more of a sport to some). The more I read, the more clueless I find myself. That's a good start of course; knowing what you don't know.
I seem to be running into some problems and I really could use your help. Please give it to me straight, I don't need sugarcoating. However, I deny throwing this jar out whatever you tell me.
 Substrate: Rice (unground), coffee, straw, gypsum. Verm layer of about 1 inch. Treatment: after full colonisation, took the lid off, poored water inside and let it soak for 4 hours, the remainder was poored out. Then I put the jar inside a ziplock bag with a little water at the bottom. The top of the ziplock is opened slightly, I mist 3-4 times a day, 5-10 times a day I replace the air inside the bag completely. Strain: 'Amazonian' Temperatures: fluctuate between 20-27 degrees C (68F-80.6F) Light: well lit room Time for colonisation: 2 months Time after 'birthing': 3 weeks.
Issues: No more colonisation of verm layer // no visible growth in verm layer. But I think I saw pinning.
Questions: What the hell am I supposed to do!? Do I mist too much? Am I just an impatient MF? Frankly, I just want you to hold my hand and tell me what the hell I am supposed to see here, and what I can do with it to improve.
I don't know if I over watered it, or if it's too dry. This is my first try with cubes that form shrooms instead of stones. I really can't place the 'symptoms' to a cause. Are these even symptoms?!

 Hyphal knotting is my guess. And some bruising or trich formation. Time will tell which it is. My money is on trich. But that's not the problem.

I don't really see any pins yet.
But it doesn't look too bad.
Are you trying to fruit invitro or what?
At any rate if you are fruiting it it needs FAE and it won't do much in that sealed up bag.

Thanks for your response!
I am trying to fruit somewhat in vitro. The (partially open!) ziplock bag is my 'terrarium', the dry verm that I mist a few times every day is my casing layer and the jar contains my substrate.
I open the bag at least 5 times a day completely and replace all air inside of it that way.

For your next batch- try PFTek, it's the easiest- and you would have been able to birth the cake if you used wide mouth jars. Look up SGFC as well (shot gun fruiting chamber)- having your sub still in the jar severely limits its FAE and humidity.

I am on that as we speak TravelAgency, Wide mouth jars of Azurescens, Z-strain, B+, Amazonian, Reishi, Oyster and Shiitake are now colonizing (I get a little too enthousiastic sometimes). I have been battling contams since day 1, and it's getting better after throwing out and preparing a couple of jars every week. The same goes with techniques. I like practising and experimenting It's just that I never got this far.. I wanted to see if this technique would work as well. Since the 'V-tek' uses a similar method (as far as I understood; fruiting from inside the jar) I figured I might give it a try. I can always pull it out of the jar if it keeps being stalled for a longer period of time. I just don't know if I'm on the right track.
I have a SGFC with a B+ cake that was heavily contamed. So I use that one as an experiment to see how things go - and shoudln't go! - in a SGFC. Of course I will clean it out before I try my next cakes.

My suspicion was spot on. Trich took over. Jar is emptied out in the backyard.
