Stereo Microscope Advice
    #21807390 - 06/14/15 05:50 PM (8 years, 7 months ago)

I am looking to buy a stereo microscope and have come across this old Nikon SMZ-2 for £180 ($217). It comes with x10 eyepieces (stats are in photo below).

How is the pricing on this? I'm not experienced with microscopes and the second hand market in the UK is practically non existant so finding it hard to compare prices.

Any advice on this is appreciated.

Thanks!

Here's a picture and info from Nikon catalogue

This is actually a dissecting scope, used to look at "larger" scale things. So it would be ok if you wanted to see, say, hyphal patterns and such in agar. If you wanted to look at finer things, such as spore or basidia morphology, you would want a compound scope (which unfortunately cost quite a bit more).

That said, this scope would cover you fairly well for many purposes. It can see spores at its higher magnifications, so you could verifying some things like spore germination or the like. I use a similar one in the lab for single spore isolations and hyphal tipping.

(EDIT: And I completely missed that you specified you were looking for a stereo scope. In that regard, yeah this guy is good, and not a bad price. You can get a less brand for about 150$, but nikon is a good name)

In terms of a decent compound scope you can find some good starter scopes on Amazon in the 200-250$ range.

http://www.amazon.com/AmScope-B120C-E1-Magnification-Illumination-Double-Layer/dp/B009VUPIKM/ref=sr_1_3?ie=UTF8&qid=1434327035&sr=8-3&keywords=microscope

They arent top of the line by any means, but they're plenty good for a gifted amateur or curious student.

Thanks for the reply. I think I'm going to go for it and get the Nikon one. Just to double check, if I wanted to try my hand at crossing strains and hybridising then this microscope would do the job?

Sure, at its higher magnifications you can pick out individual spores with relative ease. They're tiny, but still visible. I had do to do single spore isolations all the time in lab.

You streak some spores out (fairly dilute) onto agar, wait till they just start to germinate, then use the scope and a syringe needle to cut individual spores out. place them together on your surface of choice, and if compatible they'll fuse and form a happy dikaryon.
