Where to transfer from (cube and atl7 on agar)
    #21804108 - 06/13/15 10:49 PM (8 years, 7 months ago)

I transfered tissue from a b+ fruit body a while ago and all 10 transfers came out clean so far but I can't find sectoring in any and all plates are showing complete tomentose mycellium.  Where should I grab a sample?

Also I'm new to atl7 mycelium.  These plates are one transfer after spores were put on a plate.  Anything stand out that I should transfer?

In the first picture I would transfer the two sections at 3 and 8 o clock.

On the ATL I would do 12 and a piece from the strong but slower growing 8 o clock.

It should be fine just don't touch anything from 9-2 o clock and transfer small pieces.

Thanks for the responses.
so the 2 plates shown were rendered useless because of old parafilm I used didn't hold up.  Picked the plate up and the lid came off so I took the cover off to take photos.

What about those positions make those areas the best to transfer so I can infer which areas to take from my other plates?  Any particular reason these plates turned out like they did?

When theres no clear sectoring and especially on the initial agar transfer, i just pick a few spots at random
Then on your second plates the sectors should be pretty obvious

I dont think its nessecary to dump plates just because the parafilm came loose tho

:whathesaid:

Just pick the best looking growth.  After tranfering a couple times you won't get much sectoring.  It just take doing it a few times to able to spot what you think is best,  no one can teach it.
They both look good to me with the exception of the fuzz on the first picture at the top,  it could be mold or it could just be weak.

It didn't just come loose, the cover (top half of plate) Came off the base and exposed the plate to open air.

Alright I'll get those done today or tomorrow.
thanks for the help everyone
