Easy agar instructions

Ok so im tired of shooting spores in a jar an hoping it works. Agar is a lil hard to understand as i dont have much time to search. I need a step by step tek from start to finish. How to inoculate how to take a wedge and how to not fuck up haha got yeast infections from my g2g jars and before that i wasnt sure my master jar was up to par. Please help me. Also i wanna try to avoid ordering petri dishes but will if i have to. Thanks again

Well if you want to do this hobby, you should be willing AND able to research lots of things. Do your homework, no ones gonna feed you by hand.

they're the same thing, but one uses the supplies he most likely has right now.
Edit: never mind. I'd go with what he said. The brf one is alright, but I don't like how it says inject through the polyfil. What I did my first time was the agar substitute tek, but I used a jar with a sfd, and then just opened it in a sab and squirted a spore solution in the middle of the medium. I think that's basically pasty's tek, so go with that one

They're not the same. One is better. It says to PC agar for 15 mins From '04 huh?

Yeah my bad. I followed it a while back, but forgot that I changed it up a lot. Basically I just did pasty's with brf instead when I first started

Pouring in a SAB ? Seems like more of a hassle than its worth, i always pour my agar in the open, as hot as possible After pouring i let them sit for a few days and in the rare event a contam shows up i just toss it
I dont see many people using glass dishes anymore tho, i guess contams are more annoying when you use the disposable plastic kind

idk, i guess it depends on the general condition and climate of your workspace, i have a SAB but just setting it up would take me longer than whatever agar work i had that day Some day i just tried it without and it worked fine

I suspect that just means you need more time / experience in a SAB.
After a while you bond with your SAB

Ok i have been reading alot an i do apologize for the noobness on this subject, in the past ms syringe into wbs worked for me great but steadily started to lose yield and contam problems arose. Granted what i would do is one master jar with the syringe let it colonize g2g 10 with that jar an so on. But since its a shot in the dark with the ms syringe i have embarked upon the agar journey.
I have copied an read pasty's agar tek an only place i am lost at is inoculation. Is it threw the micro tape? Do you just undo the lid an drop a few drops from the syringe? (In a sab of course) i suppose with a petri you would have to right?
Planning on getting petri dishes an some gt syringes next week
Also wtf is pouring what pours where lol thank you for the time save!!!!!!

With pastys tek you fill up the jars with agar, then PC (iirc) When you use petris, you PC a big jar of agar and then pour it into your dishes I figure if you pour it while the agar is as hot as possible, right out of the PC, any airborne contams that get inside your plates are destroyed Problem is when those plates cool down they suck in outside air, so some of my dishes get contaminated at the edge This can be avoided by using parafilm, but that shit is expensive and personally i just find it much more convenient to pour a few more plates, toss the 10%-20% contams and just put my glass petris in the dishwasher
I make a big jar full of agar than can be reliquefied over and over and then it takes me about 5 minutes to load the PC with a dozen plates and the agar, let it sterilize everything and then another 10 mins to pour all the plates on a clean countertop Just setting up the SAB, cleaning it thoroughly, loading everything in there and cleaning up after is more than half an hour
Pastys tek is the most foolproof tho, this just happens to work for me because theres very clean air around here, kitchen is brand new and the entire house is solid brick If you have lots of drywall and wood, i imagine its trich city

Pouring is pouring agar into the sterile pitre dishes.
Yes when you are doing agar work you are going to have to learm proper sterile technique so that you can open your plates to inoculate, make transfers, cut wedges etc.
I find its better if using a syringe to drop onto a sterile cotton swab or a loop and swipe onto your blank agar plate, this makes it easier to pick clean growth because you have an idea where the germination should be. Just dropping spores directly onto the plate the liquid rolls around amd spores germinate all over the plsce, can grow over contamz

Thanks everyone mucho appreciated, definitely cleared up alot of mystery.
Thoraxx thats what im talking about. Im gonna do the easy plates for my noob trial run and the order some glass plates, have read both good and bad about them but i love a re-usable item.
House is full of carpet and is a trailer so contams in the air are 100% everything in a sab.
Can someone give me a slap in the face if my method is wrong when using my sab. 1. Hepa filter room for one hour with everything in room. (Jars, sab, assorted items) 2. Shower while filtering air 3. Adorn the tyvek suit and begin wiping down jars with iso, i usually get soapy water and try to coat every inch of my sab with suds. 4. After sab is wiped with soapy water i wipe jars again and place inside master jar last. Then i iso myself and get in.. My problem is that when i load jars im constantly opening the sab to place them in.... Go ahead and laugh cause i do half the time. Any tips?

if you want to use a hepa make sure to turn the fan off 30 minutes to an hour before you start work in the still air box.
you really don't need to go through all that trouble. just close all doors and windows, turn off fans or central air.
put everything in your sab and spray your box down with everything already in it. wash up, put on your gloves or whatever else you want to wear, iso and get in. wipe down everything inside the sab with 70% iso and do what you need to do. flame sterilize outside the box because warm air causes currents and thats the opposite of what you want.
work quickly but smoothly and you'll be just fine.

Right on cool so thats not a major factor lol. Trying to eliminate every angle of contam,
My guess is spawn fucked up due to ms syringe. Another tub contamed as soon as i fruited, jars fully colonized mixed to coir an verm pasteurized for a hour an set over night but nerp after three weeks colonizing i have two pins an a huge patch of bubbly white green shit.... Sigh.... I see pics on hear an wonder if i ever really did grow lmao
