Fuck it all to hell. I just saw green in a bag, again. I don't know how to be any more careful, I really don't. I have gone over and over my technique.

The bags are run long enough to kill mold so that's not it. My bags are opened the shortest amount I can get away with and inoculated with what appears to be 100% clean wedges.

I've tested my hood by opening a plate, facing it towards the filter and moving it over every inch so if there was a leak it would be easily detected, the plate stayed clean.

This used to be a fun hobby, used to. Now I'm all fucking stressed out because everything I do I have to throw away.

I'm out.

That's nice dear.

Maybe stop using bags?

still air box, pp5, whole brown rice, LI/LC, coir/verm.

You seem to have been over everything, but you are clearly missing something. Try smaller jars (pints or even half pints) and noc up a shit ton of them... Keep it simple til you get the results you are looking for.

I didn't have any better results with jars. It's either something I'm mistaking for mycelium on my plates or I'm just ignorant.

It just happened one day out of the blue and now I can't grow shit.

How much success have you had with bags in the past? I suggest switch to jars for a single grow. That way you at least narrow the vector.

I had very limited success with jars before I switched if you remember.

But I could never nail down the cause. It would kinda come and go but I'm not the best at tracking my cultures. Although I scrapped every culture I was working with and went back to spore->agar.

pc issue? Have you left jars or bags sealed after pc'ing to see if they get contaminates growing in them?

If it isn't your grain prep, your sterilization, or your culture then its how you are inoculating. It has to be one of those. That's why I suggested lots of tiny jars. You can test a ton of variables in a short time.

It's either something I'm missing on my plates or my technique.

I dunno what else to do for my technique. I've worked commercially in a hood and am quite familiar with usp 797 and aseptic techniques.

I wanna say its not my technique since all of my agar work is clean ie: no bacteria or colonies

But I wanna say its not my cultures because I always use agar and it always looks exceptional.

Fucked if I know

Even if your technique was shit, you'd probably not have as many problems as you do. I g2g using ziplocks in a sab, fer fucks sake and don't have contam issues at all.

I doubt you could have so many crap cultures, though... clean growth over a few transfers is enough. Now I'm stuck. Wanted to help you troubleshoot, but sorry buddy.

yup, switch to jars and see if that helps.

and run through your sterile technique to make sure you arent using any short cuts (such as reusing old gloves)

Try some PF cakes. Dry everything in the oven to be sure it's all at the perfect water content when you hydrate.

If those work it's your grain prep.

First one looks good, second has a spot of bacteria at about 8 o'clock.

The second plate came from the leading edge of the first plate before "it" grew out.

Yeah neither are good. The second plate has a piece I could grab from but this is what is piggybacking all my cultures and I cannot seem to get rid of it.
