Incidentally, look what 23andMe did for me a while back for just $100. It's genotyping, not sequencing, but still pretty friggin awesome. :rockon:

I think genotyping would require sequencing wouldnt it?

Nah, except for telling me I have brown eyes, curly hair and psoriasis, which I already know. :grin: I'm not sure it's accurate enough to take seriously when it comes to health planning or medical treatment, although they're incorporating new studies all the time. But I'm not really good about taking care of my health to begin with... a brisk walk four times a week would probably do me a lot more good than all the genetic screening in the world. Mostly I just got it for entertainment purposes, and a morbid curiosity to see how I might die.

The genealogy part of the site seems good for getting in touch with long-lost relatives though. I don't have much interest in that, but I have an inbox full of like 80 people who are likely to be fifth cousins or closer and want to compare family trees.

Pscientist, I'm kind of confused about the terminology too and I might not be using it right but they do answer this question at https://www.23andme.com/you/faqwin/sequencing/.

Yeah, all that is doing is extracting and running DNA on a gel. Bc you are not cutting it with any restriction enzymes, you couldn't tell one person from another... so it really isn't fingerprinting. Although they acknowledge that at the end, saying that the only thing you could do with that is tell apart different species.

On the subject of DNA sequencing, however, it shouldn't be too many more years b4 a company comes out with a bench-top size sequencer (process will be called 3rd generation sequencing I believe) that can sequence an entire organism in less than an hour with a recurring costs of under $100 (basically the reagents.)

The method is to utilize fluorescence molecules that will be attached to every nucleotide in the solution, so that each addition (via the polymerase) will generate a different color that will be picked up by a detector in real-time. I heard a talk on it last summer by someone at our university; I think I remembering it correctly more or less.

Prior to sequencing I think that aliquots of the genomic DNA get cut up separately with a few different REs so you are sequencing multiple pieces in separate reaction cells, & then the software puts them together by matching overlapping sequences.

Whatever company first comes out with such a machine will do very well. I'm inclined to say that such a machine will be very expensive, but apparently various companies (that anticipate they will be marketing such an item) are making claims that it won't be all that expense... so we'll see.
