6 D-size and 3 9-volt batteries... After a few months of batteries you could have purchased a regular centrifuge.

You are kidding right? It's a neat kids' toy, but it doesn't sequence real DNA. You need TOPO cloning stuff or whatever to do that and a $225,000 sequencer. We don't even have a sequencer -- we just clone genes into TOPO and then send them to a company to have it done.

It does DNA mapping, and it is real and will work with real DNA... by what the literature says.

We did in High School. And I guarantee we didnt have no 225K machine.

Yeah, how? No, you didn't sequence DNA in high school. I only do this stuff for a living, people -- I think I have some idea about how it works.... I mean for fuck's sake -- you cannot sequence DNA without really expensive equipment.

BTW -- the electophoresis is fake, too, because you need mutagenic dyes to stain DNA and high-powered UV light.

Sorry to let your hopes down, kids....

EDIT: It is pretty misleading, though.

Yeah it's a toy, to seperate DNA you need an far more powerful centrifuge than than that. If it was real the main safety warning wouldn't be "This product contains small parts that may present a choking hazard for young children".

people have made homemade PCR machines for cheap. Yes, it used dangerous expensive chemicals.

What make commercial DNA sequencing so expensive is that you would need hundreds or thousands of devices similar to this in an array compiling data, and acting autonomously.

Actually I was wrong -- I checked the list prices just for fun and sequencers go for as low as $98,000.

A PCR machine is just a thermal cycler -- it just changes temperatures 3 times per cycle -- a denaturing temperature (breaks the ladder), an annealing temperature (where random base pairs that you add anneal to each seperated rung of the ladder) and an extension (kind of like an error preventing step to make sure all the DNA is complete.) A thermal cycler could be done by hand with 3 different water baths at different temperatures.

You'd still need the enzyme, though (polymerase) a buffer for the enzyme, DNTP's (base pairs, A,G,T+C) and primers to hook to one part of the sequence.

Most sequencing is done by taking out a gene using specific primers (a DNA sequence to start the PCR out that the enzyme binds to) but you need primers for this, which means you already need the sequence of DNA.

The easiest way (IMO) to sequence unknown DNA is to use these things called transposons that are like mobile DNA, and they bind to random parts of the genome. You can then take these out and clone them into cells (usually E. coli) and each colony will have a different part of the genome. These will have overlapping sections, and you can put all of this into a computer program which fills in the puzzle. With enough of these you can map the genome of anything.

It doesn't cost a lot of money per sample, but the cloning kits cost a lot, and altogether you have to invest at least $10,000 because you go through a lot of equipment.

How much of this is to cover product liability? Remember when this is done on a professional level,people's lives can be at stake. I don't doubt that the toy works,but can you trust the results?
