Agar to grain questions

Hey guys! So I have a few questions to about doing agar to grain transfers.
First of all, how big does the wedge need to be to transfer to a quart of grain? How exactly do I pick up the wedge? Like do I stab it with the scalpel?
And how long should I expect colonization times to be?

You can use a whole plate or a rice sized piece. Colonization depends on too many things.
And yes, stab that mofo like it humped yer gal. In reality its kinda a stab and scoop motion but you know, whatever.

Yea what madchef said. Just continue to be as sterile as possible. If you can try different wedge sizes and date your jars so you can see how long it takes. Fun little experiment.

Using a piece of culture the size of a grain of rice's place is warranted when doing agar to agar transfers i.e. isolating, when cleaning dirty cultures, cloning. Agar to grain transfers warrants the use of the "wedge & transfer" technique.
The more wedges that you place into a single jar will dictate colonization speed in the favor of faster colonization. Administering a mild & gentle shake upon inoculation of your agar wedges creates more points of inoculation by breaking up the mycelial culture, and spreading live culture through out the grains evenly expediting colonization, as well.

For my current run, I used a single agar plate which was nearly colonized to the edge, and scored it into 8 sections with the tip of a sterilized 45-degree angled probe. Then I used that probe to lift each wedge (1/8th of a plate) into a jar. I really just wanted to see how much faster things would go with a wedge that big instead of the little 0.25 x 0.25 inch chunk I was using previously. Turns out, it's a big difference for not a lot more work in maintaining agar plates. This run of jars is at the 30% colonization mark after 8 days, and they're ready for a shake.

I also usually use several wedges per jar when I have them. It really depends on how many plates I have. The more the better IMO. Getting ready to do some myself.
