Grain to agar first time

SO I had four decent looking jars of spawn fully colonized with a few of those pretty stray grains that stuck high up in the glass that were also colonized with a lot of aerial myc. Yummy.

It was from a spore syringe I got from a buddy. I hadn't thought about it before but it hit me that those stray grains would be perfect to drop on agar. Used standard ops with a scalpel to transfer. I completed 12 plates.
My camera will not focus past the top of the petri lid very well for some reason.
2 days after transfer

4 days after (yesterday)
 3 days after (yesterday)

I would have transfered them yesterday to clean plates but I was exhausted from work. I'll be getting that done today. It seems I got very few sectors from these stray grains upon inspection of the bottom of petris.
Jars were spawned to hpoo and straw substrates same day as transfer as well. Feels like I really got the most out of these jars

Nice job...some clean growth going on. You should definitely do some transfers. I'm no agar expert so I'm sure other can chime in but I think I see a sector or 2.

Oh yeah there are definitely sectors but not as many as I would have guessed from spores. Hopefully after this transfer I might can get an isolate that's clean. I have to make a move soon so I'm scrambling to get what little of a culture collection I can so I don't have to start fresh again.
Here is a transfer from atl7 sclerotia I grew invitro on a no pour plate.

3 days later

As soon as I clean it up and isolate it's going straight to slants since I don't have any time to mess with right now.
I got some Pan Cyans on agar too but they are going slow and weak. Any suggestions anyone?

Might take a few more transfers than that, from spore usually takes 7-10 transfer before I even see sectoring
Your end goal is one isolate yes but thats after youve tested them all. Definitly isolate all the sectors, so you end up with 8-10 isolates. You will have to test each one under the same conditions, and pick the winner for yeild and (dont forget this one) potency. So maybe dont slant yet, just store your round of isolates on plates in the fridge untill they are tested
