1st Time Grower - Stalled Jars
    #21780587 - 06/08/15 08:23 PM (8 years, 7 months ago)

SWIM is cultivating P.cubensis B+ for first time.

Using YouTube videos and the book, "Psilocybin Mushroom Handbook" by L.G. Nicholas  Kerry Ogamé, SWIM followed the PF Tek instructions to the letter.

SWIM found there to be a lot of conflicting information about "do's" and "don'ts" with regard to to various aspects when doing research online.

SWIM successfully inoculated 13 jars and started incubation process. Mycellium started showing within a week, but took like 3 weeks to fully colonize the first two jars.

Following given instructions, SWIM kept incubation temperature around 86F. But in retrospect, SWIM found tons of  conflicting information about temperature. Regardless, the mycelium grew (though very slowly).

SWIM moved 2 jars from incubation chamber to birthing chamber after 40 days -- they have now been in birthing chamber a total of 23 days with no sign of primordia.

SWIM did additional research while waiting for the remaining jars to colonize. SWIM discovered the "dunk and roll" technique.

After 62 days, SWIM dunked 11 cakes for 24 hours in cold water, completely submerging them. At day 63, SWIM rolled them in dry vermiculite and placed in birthing chamber.

The birthing chamber parameters are as follows:

-Plastic bin with about 20 quarter-inch holes punched in sides to allow for FAE.
-Saturated perlite to keep air moist. RH is at a constant 99%.
-Reptile mat adhered to side of chamber, keeping internal temp between 70F and 75F.
-Fanning carbon dioxide out of tank 5-6 times daily. Misting cakes with spray bottle at same time as fanning (after).
-Lit with fluorescent light for 12 hours a day. (Though some only do it 8 hours.)

SWIM is really perplexed that she has followed all the instructions -- even though some of these instructions were contradicted depending on where SWIM got them from.

I guess SWIM's questions are as follows:

1. Is it possible the high temperature during incubation could have dried the cakes out and permanently damaged the mycelium growth?

2. Can a "dunk and roll" effectively save cakes that might have become too dry and re-hydrate them?

3. Is this a complete wash? Should SWIM just try to start over?

4. In general, if one does everything the right way (which SWIM is realizing is a completely relative term), how long after being placed in the birthing chamber in the most ideal circumstances, should it take for the cakes to show signs of growth?

SWIM is really at the end of her rope. Over two months into the cultivation process, and SWIM doesn't seem any closer to the end.

SWIM really needs your HELP!

Beforehand: We don't SWIM on here. Using SWIM is a hell of a lot more suspicious that just saying you're doing it. Most of us just read it as "Someone Who Is Me"

1: 86 is on the high side. The "Golden Zone" of colonizing temps for jars, regarding cubes, is 65F-75F. Going up into the mid 80's will allow some competing bacteria idea temps to grow. I don't think the temps killed your jars, just colonize at room temps in the future.

2: Yes, though my understanding is that the "roll" is only done after the initial dunk, and is not necessary on future soaks.

3: No, and no. Plug up your holes with polyfil stuffing, tight in the bottom holes, loosely in the top ones. And only fan after each mist, it's not a "times per day" thing. Misting is the same way. Only mist when the sub looks dry (ei: not glistening), and fan briefly to disperse the new moisture in the air.

4: Patience - rule number 1 in this hobby. I'm going to guess this was an MS injection. Multi-spore inoculation is a turkey shoot, you never quite know what'll happen. Some jars take off flying into colonization, others take a month or more to birth.

All in all though, if you could upload a few pics of what's happening, we'd be much better able to asses what's going on. Until then, this is about the best advice I can offer.

Hope it was helpful

I agree that this is solid advice however the OP seems to be going for a Shotgun, not a mono. There is no need to stuff polyfil in his holes.

OP a pic of your setup would make trouble shooting easier.

A proper shotgun should have 1/4" holes spaced 2" apart on all 6 sides.  Make sure your perlite is at least 4-5" deep.  Use a few blocks of wood or whatever under each corner of your fruiting chamber to raise it up a bit off the ground. Air needs to be able to freely circulate around all 6 sides for it to work properly.

My apologies, I'm so used to monos lol.

OP: Disregard my polyfil comment. Everything else stands though. I'm also curious why you have a heater in the tank? Most monos and SGFCs don't need a heater and work off ambient room heat. What temps does your grow room reach during the day/night?
