After shaking 8 quart jars and 4 didn't recover and colonize, after 5 days of waiting and scrutinizing I took them out in my garage and cracked them individually and they smelled sour, yuck and toss. Ran through dish washer and started another 4. Luckily this time rather than ms I've been working with agar and have isolates. Round 2 slut contams.

Sounds like your contam issue came from improper sterilization of media, and/or overly hydrated grain content, and/or dirty inoculm, and/or colonizing/incubating at too high a temperature range.

You should give more detail about the event leading up to the contamination. What substrate, how did you sterilize it and in what, method of inculation, sterile technique, where the jars were store after inoculation and at what temperature etc... With more information, what may not be immediately apparent to you may be obvious to someone reading your post. Don't give up cause you can do it! Good luck!

Rye grass seed I sterilized in a PC and nocced in a still air box, I think my substrate was too wet. It was noticeably very moist in the containers, overly moist. So I put the grains out on newspaper under a fan, then loaded 4 quart jars today and pc'd at 15-16 psi for 60 mins. Letting them cool gradually in pc as we speak

So this is a new attempt? What are you growing on the seed? And how long did you PC the see and at what PSI?

Just to let you know ham is most likely right. Grains have endospores that seem to be barely able to survive 2 hours in the pc at 15 psi. 1 hour isn't enough.. 90 minutes isn't enough in my opinion.
Moisture content is also critical.
