Cloning question. Use shroom or Mycelium?

So I've got some agra mixed up and want to get some practice isolating a strain.
I've only got about 3.5 grams dry per flush from my 2, 1/4 pint cakes. So I don't have alot to choice from. lol.
Anyway My plan was to take a sample of the "fuzzy feet" part of the shroom. Which is mostly Mycelium.
Can I do this?
Most harvest the shrooms and take core samples to preserve cleanliness but I plan to move this through at least 3 agrar plates anyway so I have no need to get a clean inital sample.
Is taking th Fuzzy Mycelium ok?
My goal is to get a clean agar wedge with a guaranteed fruiting strain and get it into 3 pints of WBS by next Tuesday.

That's a good way to see bacteria take the entire plate over. Bacteria can be way faster than mycelium so that's why we try to get a clean spot.

Your best bet is to sanitize an exacto knife and cut the shroom in half long ways. Extract a sliver of shroom core to the agar. Do this in a still airbox. Myc is most likely to be killed by contams in its early stages, when its network isnt strong yet.

^^^^^^^^^ Sorry, but that's bad advice.
Don't cut the stem in half with a blade. A sure way to bring contaminates from the surface to the core. Tear it in half, to expose the center, then use your flame sterilized blad to take your sample.

split the stem in half from the base with your fingers and take a meaty sample from the base of the stem. I get a lot more clean samples from the base because it's so big. Also you can just wait until they're ready to pick to take a clone. Most recommend a pin but I personally do full growns. In the wild we take samples of full growns too, so its not much different.

Ok, I went ahead with the clean approach from the stem. What I did was cut off the shroom base and dip it in 3% hydrogen peroxide for about 5 min.
It got very fizzy.
Was this a bad move? Did I kill the Mycelium by doing this? I read about others prepping their clone by doing a H202 dip.
Like I said I'm practicing for when I need to do this for real.

You don't need to do that. Just pluck a shroom from your cake, bring it into your SAB, rip it slightly with your fingers, take a biopsy from the center with your flame sterilized blade. Your chances of getting a clean sample is pretty good 
It's excellent that you're dry practicing though. Great idea

I'll make more Agar plates tomorrow.
Also my X-acto knife kit didnt show up yet from Amazon. ( bought it like Thursday of last week, wtf) so I used a normal knife sprayed with 70%iso.
Whatever Like I said this first batch is practice. I'll get some agar for WBS started next week with the real deal.

Yea wiping your blade with alcohol wasn't the best option. Look up the difference between sterilization and sanitization. You just sanitized your blade, not sterilized. Use a flame until the blade turns red hot. Blue flames are hotter than standard yellow.
Sanitized = with chemicals. good Sterilized = with heat. much better

Usually you would pick a fast growing pin from a big cluster but since you dont have much choice just take the best that you have.
But if I were youd Id do a few bigger MS grows do I could search out some really good clones!
Lately Ive been taking my clone tissue from the inside of the cap, the tissue is softer so its easier to drag the blade and get a nice biopsy and also its been said by some trusted members that you are more likely to get a single sector culture from this area

Interesting. I never heard about that before. How has it been working out for you?
You avoid the underside of the cap because of spores, right? So what do you do? Slice a piece off the top of the cap to expose the flesh, flame your blade again, then scrape?

Ive only had a few chances to try it so far so I cant vouch for that yet. But it definitly is easier to grab a piece of tissue from the stem can be kinda tough and stringy inside
Yea you do wanna stay away from where any spores would be. I tear the cap with my hands and drag the sterile blade along to grab a piece of tissue
