Sterilizing synthetic filter discs
    #21718171 - 05/24/15 04:58 PM (8 years, 8 months ago)

I've got a contamination that keeps coming back every batch I do but what's strange is none of the agar plates I do contaminate
I normally just wash out the old bottles in the sink fill them up with grain pressure cooker for 90 minutes and I'm done
but I'm thinking there's a contaminant that lives in the filter discs
My next batch I'm going to soak the bottles and rings in bleach water over night, wash them in dishwasher and soak the rye for 24 hours before it cooks
But what could I do to sterilize the synthetic filter discs

A few questions. What's your inoculation method? What's your contamination? And what kind of filter discs do you use?

I've been inloculated with syringes and agar they both produce the same results
This is the contaminated bottle of rye

I use the synthetic filter disks 90mm from fungi perfect

Do you know what bacteria smells like? It looks like it's covered in it. I think you need to up the sterilization times. It always seems to be better if you do. You want to kill as much bacteria as you can. We have to pressure cook for bacteria. If we didn't need to we could just get away with a boil for half an hour or just a typical pasteurization. I assume you flame sterilize with agar?

Yes I flame sterilize everything I can as well as a 10% blech spray for all the surfaces in the flow hood
Normally a 90 minute Cook in the pressure cooker for the bottles of rye

Yeah like I said. Minimum should be 2 hours.

90 minutes
120 minutes.

Altho lots of other things could be a factor. It seems to be a big consensus all around that upping the pc times really helps.
