That is a fine way, also cuts the genetics down real fast
Thanks, everyone, for all the good suggestions. This is why I keep coming back.

You want to make a streak with the droplet using a cotton swab or inoculation loop. Don't just squirt a drop in the middle and leave it or it's incredibly hard to isolate from any contaminants that are present. I usually just make the streak in a circle around the petri.
Hopefully even if there's bacteria you'll be able isolate at least one good colony.

That's probably nailed down much of my problem right there, is that I've done just what you said not to; leave a small droplet to sit in the center of the plate. So, do you do a typical T-streak?

You can streak it how you want but make it one long streak. If you have a large petri you could even zig-zag it. Around the end of the streak where very little inoculant is left and colonies tend to be sparse and spaced apart is where you'll most likely find your usuable colonies. It can be very difficult when dealing with bacteria though and antibiotic agar can be extremely helpful.

I did up 4x 1/2pt jars along with the quart masters I PC'd earlier this week, and plunked a fairly heavy spore load into them. 2x Treasure Coast and 2x Golden Teacher (both some of my favourite looking cubes) Only 5 days in and I'm already seeing growth in all 4. When I say heavy, we're talking 4 big hanging drops, then I shook the heck out of them. Looks like I'll be ready to transfer out to agar in the next few days.
Thanks again, everyone, for the suggestions on fixing this issue. I can hardly wait to get these guys cleaned (if needed), ramped up and ready to go.
