MS grow to strain isolation

I'm planning on doing a monotub grow from a MS syringe. Once I grow out the fruits, how do I isolate a culture? should I just pick the biggest fruit and print it or clone it on agar? after that, will i need to isolate it further? a little help here please. thanks

pick a small pin from a cluster and clone that on agar. some of the links in my sig may be helpful

You would wanna take a pin from a fast growing culture, or any other mushroom with desirable traits to clone. This will be a clone, you can isolate sectors from it if you like and test them out but it will still be a clone.
If you are after an isolate, the process is essentially to keep making transfers away from your germination plate untill you see defined sectoring, then isolate each sector to a new plate untill they no longer show any sectoring... then test out each isolate and pick the winner.
Both are great ways to get great cultures!

Not necessarily looking for an isolate. Just want to "selective breed" a MS grow to achieve a repeatable, abundant grow. so should I clone the biggest fruits and groe out those? To clarify, how should I achieve genetics to give me the most repeatable, abundant grows? thanks

If you clone once there are still gonna be variable characteristics in growth patterns...it's not really an identical clone... he is talking about sectoring the different growing pattern of the mycellium until you have only one style of growth that is agreessive (or whatever characteristics your searching for)...so pretty much if your gonna clone you might as well continue to isolate... I don't know if that helped...

Take your favorite shroom and clone that one, or clone a pin from the cluster if there is one.
You can keep taking clones from the next generation to get closer to your desired traits. Since you are starting from MS, the genetics will not be the same in every generation.

So if biggest is my favorite, then keep cloning the largest fruit? I'm just wondering about productivity. Like how to achieve the best flushes. Which I know isn't entirely genetics, but it does play a part.

Grow for potency not size. Both is great if you can get it haha.
I'd suggest you take many clones, then sample them. You don't know how potent they are until you eat them, and by then there's no shroom to take a clone of lol. So you have to make clones before you know how potent they are.

Genetics are probably the most important factor in how a strain grows. Poor grow conditions will of course diminish yields though.

Lots of testing and searching for winners.
Im in the process of testing out lots of AA clomes Id taken. See the tubs in my sig? Those were clones taken from great clusters, I still have a lot of them to test but so far the three tubs that fruited already were pretty awesome. I still have to try them out to test potency.
Essentially Im going to pick the very best out of the best, and slant it.
But yea, at that point you could continue to work on the clone, isolating out each sector, then fruiting and testing out each one again.. but most of the time the clone is fine as is.
