Spores germinate in one place if you put them in one place... Doesnt have much to do with it though
So it seems...
You should try to avoid that, it'll colonize quicker.

Rhizo/tementose means nothing. I've had pure cultures go back and forth depending on the concentration of food.
This method of using MS bags would work. But it wouldn't be 100% consistent. I want to be able to know what my yield will be.
There's also the variable of spores being dirty. I'd expect 10% contamination with this method. That's way higher than the 1% I allow.

But it does mean something. It's indicating a change. Now in a situation where the substrate isn't altered and you see it lose rhizomorphic growth what do you thinks causing it?

From my experience it seemed to have a lot to do with humidity.
That's bro-science though. I got nothing to back it up.

I'm a little late to the party but ....
Micro, what sort of sterilizer/pressure cooker are you working with?
In my mind, the Prestos and the AAs do suck (unsterile) air back into the pressure cooker as they cool. If they did not, there would necessarily be a vacuum inside the pressure cooker and the lid would be near impossible to get off(I can explain why this is so if an explanation is needed). Therefore leaving a vessel (for liquid media) lid slightly open (when pressure cooking) would create a vector for contamination. The liquid media vessel and the pc would both be cooling and sucking in unsterile air.
I'm assuming that you are sterilizing a liquid in a vessel like this right?

Let your PC cool in front of a flow hood or do as RR said and dowse a thin, clean cloth in 91% alcohol. Wouldn't those precautions help detour contamination?

Just let the filters on your jars do the work. Problem solved.

Which is what I do. I just mentioned what I mentioned because OP showed a pic of a vessel without a filter, and this idea of cracking the lid.
I appreciate the info though OP!!

if you're in a dirty area that's unadvisable. if you cool down in front of a hood then you don't need to spray the jars before opening them.

If you're in that dirty of an area, clean it up.

wanna come clean 1000sqft of grow rooms ?
I bathe in trich before I inoculate 100 bags every day, 5 days a week. It's all I can do to keep 100sq ft of lab clean.

If I had 1000 sq ft of grow space it'd be impeccabpy clean. Most of growing mushrooms is cleaning and prep work anyway.

that's why i'm changing my business model from direct marketing to wholesale. at this point I'll take a price cut if it means one 2hr delivery each week vs 10+hrs. and growing one species vs 10.

No. Neither one of those will do anything.
edit: unless you mean clean the inoculation site, then yes that will help
Why s everyone so stuck on jars? Because they are reusable?
