Agar Questions

1.) How long does it usually take for a live tissue culture to colonize agar on petri dishes?

2.) When petri dishes are finished colonizing, what is the best way to store them until use?

3.) Once the agar is finished, How much mycelium culture should be used to inoculate a pint jar of grain?

Usually takes about 2 weeks for me. You can use it before the entire thing is colonized though.

Ziplock bag in the fridge.

I use a small piece. About 1x1cm. Sometimes smaller.

i inoculate 3 jars from 1 dish because i like faster colonization times, but that's just me.

Which would finish quicker in anyone's opinion? Taking a fully colonized agar dish a taking 12 small cultures from it and inoculating 12 jars of grain, or taking say 4 larger cultures and inoculating 4 jars of grains, then taking those 4 jars when colonized and doing G2G transfers for 12 jars?

the first would be quicker - only one generation to grow.
why not grow 12 jars, then use ten as spawn, and the other two to g2g another 10 jars. perpetual motion. :cool:

In anyone's opinion, how much faster growth and how much more yield do you usually get using a pure culture strain instead of multispore?

The only difference is you get faster colonization, especially with a lot of potent inoculum (the mycelium water) and also, if you clone a really good substrain that fruits really well you'll get better results, usually, then random chance.
