Troubleshooting slow colonization

My first post on this site.  I've searched around but haven't found anything that duplicates my problem. 

I'm new at this, so I followed the PF Tek as exactly as possible. 

I have a B+ spore print about 1 inch in diameter and scraped off a little less than a quarter of it.  My syringe wielding was pretty pathetic during the first run and I ended up blowing through all 10cc by the time I had inoculated 3 jars.  I covered the jar lids with sterile foil and put the jars on the shelf. 

They receive ambient light and the room temp ranges from around the low 60s to 80 F.

It has been 2 months almost to the day and no sign of colonization, nor any contamination.



I'm fine humbly waiting, but I wonder if I didn't go wrong somewhere?  One of the jars may have a little too much moisture in it, because it collects at the bottom.  The other two look pretty ideal in the amount of moisture.  Some light precipitation on the inner glass but no collecting of fluid at the jar base.

if they haven't grow in 2 months they're not going to. the mix was probably too wet. also the foil is supposed to come off. read through the second link in my signature. follow one of those proven PF teks.

I don't know which one you followed but if it says to put the foil back on it's the wrong one.

thats a very thick dry verm layer. maybe you didn't inoculate deep enough and only got it into the dry verm. i just had a jar of chitwans take a super long time cuz i just barely got past the dry layer

Your moisture content is off because you used the whole syringe on 3 cakes. Somethin else might be brewing in there now  :morningtoke:

my jars inoculate just fine with the foil on and i don't have to worrie about contams falling through the hole

the spore solution drips down you often see growth all down the side if you go right against the glass so even if you did just the tip and didn't go balls deep you should still be ok. but using 10cc in 3 jars is an awful lot of spore solution. I would bet they were a bit on the wet side and the extra load of spore solution made it too wet.

whoever says to keep the foil on in their tek is a dumbass. it encourages contamination

how does it promote contams? just curious cuz im new to this still but all my jars have done super well with foil on. as far as i can tell i would imagine it could let contam in the holes

Take the foil off (for next try anyway, I think this one is done).  The verm layer is to protect against any contams falling through the hole.  You need gas exchange through the holes.  Use the search if you don't believe us...

who cares if contams go in the holes that's the whole fucking point of the dry verm barrier being there. the dry verm barrier is your filter. if you leave the foil on it stays moist and encourages contamination to grow on it and through it. if it dries out it works like a filter like it should. and air exchange is good so covering the holes up only hinders that too.

Thanks for the link. I looked through it and my process followed yours with the exception of taking the foil off before putting them on the shelf. 

When I did the mix it was light and fluffy with lots of air pockets in two of the jars and I agree the third may be too wet.  I'm curious, can I save these jars by taking off the foil or are the spores dead by now?

okay ill try it next time for sure and probably take em off the jars i do have going was just always worried about contams from my breath and stuff while check jars. thanks for the info though much appreciated

the spores dont "die" but I would start over if you have extra jars I guess don't have to toss these ones out yet.

It's possible, but I did my best to get the syringe deep and up against the glass.  I could see the fluid running down the insides.

I guess I don't mean dead, but are they still viable spores?  I can't see any contamination in jars.  If I take off the foil, do you think that might get the process going again or should I dump these and reuse the jars?
