So I just got done taking 3 tissue samples from a fruit and put them to agar and every piece completely blued. Are they going to work out?

Yeah. After a couple of days it'll turn completely white and really fuzzy as it starts to spread out onto the agar. Transfer as soon as you see contams. So far I've done one transfer of my current clone to get away from some trich. So far so good.

They'll be fine. Give them a few day sand they'll fluff out.
Watching clone tissue recover and crawl onto the agar is one of my favorite parts of mycology!

I was nervous too when the sample turned completely dark blue. Yours is doing the same thing mine did. It'll fluff up completely into a white ball and grow out. Make sure you transfer when you see contams the goal being not to let the contam have enough time to sporulate so you can get a clean plate.
What are you planning to do with your clone?

You want to transfer as fast as possible.

Not trying to hijack your thread or anything man so sorry if i do, but is there anyway i can do clones of tissues without agar, im in the process of wanting to get into it but trying to find the right sources. Im doing all grains too btw so could i put the clone in a plastic bag than transfer to grain after it fuzzs? Thanks guys -kushroom

Gonna grow a tote with just the cloned culture after 2-3 transfers and see what the results are, make a few master jars and possibly slant some. I'm also going to try and get down to an isolate, I bought some mini monos so I can test em before going balls out on em

Sometimes clones perform better as an intact clone rather than the sum of their parts(isolates). The different strains within the clone seem to work better in concert, rather than individually, though this is not always the case one way or the other.

Had some bacteria that turned the agar white in 2 days and started liquifying it. Smelled pretty rotten also

Yeah, when that happens I think it's time to cut your strings to that plate.
I'd hate to have that happen to a rare or important culture. Oof.

I used to do it in lc's. Grain works too. Chances of success are not as high....but....check out this thread http://www.shroomery.org/forums/showflat.php/Number/20964499

If the original clone transfers look healthy and no sign of contams should I still transfer or let the plates grow out and then just transfer to grains?
