About ready for G2G

So my KSSS, b+, and Argentina are almost complete and ready for transfer.

So here's what I'm thinking:
I have two 27qts and a 58qt monos so... 3 qt jars for the two and 4 or 5 for the other.
I have coir, MEA, petris, verm, all on the way and am excited to get clones and start isolating, so on one of the Argentina half pints, I will fruit in vitro with a casing of verm.
My jars were lookig pathetic for a loooong while until yesterday, when I bought a cool mist. I believe my jars, even though stuffed do with polyfil, are truly benefiting from this as it is not drying them out - they are uptaking water in the grain and creating their own heat. Also, rhizo growth just happened, as where before they were slowly crawling. This worked for me, that's all I'm saying, woot!

Question:
My agar supplies will be here tomorrow, so should I wait to g2g this jar tonight so I can get a strand off the side of the glass? Looks beautiful.

You can take a flame sterilized scalpel and fork a grain or two to put on plates so you can get a culture going without waiting for spores to germinate 
I just started using pints as masters too I dig it

Thank you! I have about 5 other half pinters and a qt of B+ that look just like this - fruiting them to my WBS bucket since it's empty. They look super healthy to me and almost frosty white!

Exactly what I was meaning, I wanted to get a scalpel after I pour my plates and just get a rice size strand to transfer. Off the glass. But I suppose a grain would be fine too. I just wanted to narrow it down from the get go. I'll have to do my pan cyans from agar because they have not grown at all for me using MS to WBS or even BRF substitute. Crossing my fingers.

Break the jar up and grab a grain instead of a strand, Im sure a strand would work but grain is certainly easier/prefered method IMO
I do that when I cant get germination on agar, sometimes I have better luck on grains and then I just drop a grain on a blank plate and transfer away from it

I can definitely see your point in how it would be easier as glass against a scalpel won't be very forgiving. Gotta have those surgical hands. Maybe I'll just do a few, two grains and an attempt at that strand I want. How many transfers do you typically make to get to an isolate? Seems very tedious and I'm up for the adventure. I'm starting to feel like 20 plates was only the tip of the iceberg, I can already see where I'm going to need more to continue growing clones and transfer to WBS while isolating. Will a mini fridge work at storing petris?
And yes! I'm just starting, I can't wait. I've done a few grows with monos and great yield, but always from MS. I never had time for agar nor did I feel it to be necessary as my MS was sufficient. Now I'm figuring it out with more experience under my belt. Wish I would have started sooner, it looks easy.

Isolates from spores takes a bit of work and lots of plates.
Im still learning myself, it takes me 10-14 transfers to see real distinct sectoring, and then I might take like 7 to 10 sectors to plates that each have to be fruited out and tested.
What Ive been doing is Ill make maybe 5 transfers to make sure Im working with a clean culture, take one more transfer and set it aside. Then I put the culture to grain to make clean spawn with and Ill keep working on the strain in the meantime.
But also take clones from big ol clusters from your Ms tubs, clean them up, test them out. You can also further isolate sectors from your clones too if you like, Im in the process of doing that right now actually, youll see sectoring from a clon
I work on Isolation as a side project cause it takes so long. I mostly fruit clones

That's what I had in mind, because RR says you only need a small piece as to transfer the least amount of differential genetics as possible.
