can someone tell me what i did wrong. I was making spore solution. When I made the spore print i didn't use a glove box I just put the cap in a jar and let covered with a towel for two days then allowed one day to let the moisture dry out. then i capped the jar.
When making the syringe I pressure cooked the syringe and needle for 15 minutes at 15 psi and also the distilled water. I covered the distilled water with a cap because it was in a jar and didm't use foil. Then let them sit overnight to cool
Also I forgot to flame sterilize the needle after because i thought the pressure cooker took care of that. I made the syringe in a glove box but it was a prett shitty one that i just burned holes in the side and used gorilla tape to tape the gloves.
basically that's it, now all the jars have trich

take better prints use a sab, clean foil and be carefull

Haven't you been doing this awhile? You're missing some basics on general sterile procedure, it seems.
Yeah, the pressure cooker sterilizes the syringe/needle... but then you took the syringe out of the PC and exposed the needle to the air in the room. Any sort of contaminant can now land on that needle and be introduced into your jars. This is why you flame sterilize the needle right before inoculating each jar.
