Avoid The Noid With LC's

Mycelium colonizing a liquid media. Sometime chunks of live culture float to the top. Once to the top, myce begins forming a hardened mat of mycelium. This mat grows 2 dimensionally, right over the top of the medium.

If left unchecked. The myce is allowed to fully colonize the top of the liquid media. Resulting in live culture underneath the mat to be killed due to lack of oxygen. Hard and matted liquid mycelium can often be difficult to aspire into syringes too.
Don't let this happen to you. Avoid the Noid.
Solution!:
Stir or shake your LC's.

Stirring or shaking will chop the mat up. The mat will become liquid logged and sink into the media.

Noid defeated. Repeat as needed. Thank you. Good luck!

edit: may picture taking skills have improved from this. hell, i did not even wash the jar first

PP5, plastic closures. Whatman syringe filters for GE. Soon to be modified further with large injection port. Large injection port modification for LC gun access.
No gasket. Where the lid meets the glass is triple wrapped in parafilm.

Aspire what you can of it right away. The matted myce'll be broke up enough to be aspired. Some of the matted myce will be hard to aspire still. Use 12 gauge, blunt-tipped dispensing needles.


In that situation, there's really no replacement for magnetic stirring devices. Magnetic stirring devices create a cyclone within the media that'll keep those pesky Noids under media. Until they are media logged enough to sink without the aid of being stirred magnetically.

im gonna give your grain liquid cultures a go pretty soon. im making up some plates in the next week or so to start some things off. first i have to soak some grain. its been close to 15 years since ive done that.
almost time to dust off the old stir plate and stir bars. i have those 2" stir bars.

Knock (no pun intended) the dust off that old stiplate, eats, or you're like v.
I have a thing. 1 inch stirbars go in pint size jars.

3.5cm, circular go into quart sized jars.


2 inch stirbars go into gallon sized LC media vessels.

3 attempts with gallon sized media vessels, and still no luck. It'll happen. Just gotta' keep going.

Whichever grain you use. Be sure it's end-of-the-boil-cycle grain water. EOTBC grain water has the most nutrients in it. Has a fair amount of sediment in it as well.

Great info, thanks for sharing! I gotta look for magnetic stir bars. I keep seeing comments about them and wonder if they are any better than just using marbles.

Rounding curve with LC's and magnetic stir devices. Went from stirring once a week to twice. Found the straight stir bars require a higher rpm speed to generate cyclone (1200). Circular stirbars only need 700 rpms to generate cyclone.
